Proton Nmr Spectroscopy

Proton nuclear magnetic resonance (¹H NMR) spectroscopy is an analytical method that identifies and characterizes hydrogen-containing environments in molecules, making it central to chemical structure determination. When a sample enters a strong magnetic field, proton spins align between energy states; radiofrequency radiation at resonance frequencies induces transitions, and the resulting signals reflect chemical shielding, coupling to neighboring nuclei, and relaxation. Chemists use chemical shifts, coupling patterns, and signal integration to assign functional groups, distinguish isomers, assess composition and purity, and monitor reactions. Its nondestructive nature also supports analysis of complex mixtures and biomolecules, linking molecular structure to reactivity and material behavior.

The motion regimes of ibuprofen encapsulated in β-cyclodextrin nanosponges polymer network are investigated using pulsed-field-gradient spin-echo (PGSE) NMR technique. Synthesis, purification, drug loading, implementation of the NMR pulse sequence and data analysis to work out the mean square displacement of the drug at several observation times are described in detail.

A progressive colonization procedure is described to further assess its impact on the host hepatic metabolism. Colonization is monitored non invasively by evaluating the urinary excretion of microbial co-metabolites by NMR-based metabolic profiling while hepatic metabolism is assessed by High Resolution Magic Angle Spinning (HR MAS) NMR profiling of intact biopsy.
