Elimination Kinetics

Elimination kinetics describes how a drug or other substance is removed from the body over time, a key concept for predicting exposure and safe treatment. It characterizes processes such as hepatic metabolism and renal excretion through measures including clearance, half-life, and elimination rate; in first-order kinetics, the rate depends on concentration, whereas capacity-limited, zero-order elimination removes a constant amount per unit time. In clinical pharmacology, these relationships guide dose selection, dosing intervals, therapeutic drug monitoring, and interpretation of concentration-time data, helping clinicians anticipate accumulation, adjust therapy in organ dysfunction, and reduce toxicity.

Elimination Kinetics: First-Order and Zero-Order

2025

Eliminating drugs from the body is a vital process that occurs through excretion or metabolism. Understanding the kinetics of drug elimination is crucial for drug development, dosage determination, and optimizing patient outcomes. Drug clearance depends on the rate of drug elimination and its plasma concentration. Another important parameter is a drug's half-life, which is the time required for its concentration to decrease by half. In most cases, drug clearance follows first-order kinetics,...

Kinetic Molecular Theory: Molecular Velocities, Temperature, and Kinetic Energy

The kinetic molecular theory qualitatively explains the behaviors described by the various gas laws. The postulates of this theory may be applied in a more quantitative fashion to derive these individual laws. Collectively, the molecules in a sample of gas have average kinetic energy and average speed; but individually, they move at different speeds. Molecules frequently undergo elastic collisions in which the momentum is conserved. Since the colliding molecules are deflected off at different...

Enzyme Assays and Kinetics

Enzyme kinetics describes the catalytic effects of enzymes, which are biomolecules that facilitate chemical reactions necessary for living organisms. Enzymes act on molecules, referred to as substrates, to form products. Enzyme kinetic parameters are determined via assays that directly or indirectly measure changes in substrate or product concentration over time. This video will cover the basic principles of enzyme kinetics (including rate equations) and kinetic models. The concepts governing...

Enhanced Elimination of Poison

Poison can be effectively removed from the gastrointestinal (GI) tract through various decontamination procedures. Antidotes serve a crucial role in counteracting the effects of poison by inhibiting enzymes responsible for producing harmful drug metabolites. In some cases, these toxic metabolites can be neutralized by endogenous cosubstrates, which are maintained at specific concentrations to prevent interaction with cellular macromolecules and subsequent cell death. Renal excretion is the...
