Cardiogenic Thromboembolism

Cardiogenic thromboembolism is the formation of a blood clot within the heart that travels through the circulation and obstructs a distant vessel, making it an important cause of ischemic injury. Thrombi develop when blood stasis, endothelial or endocardial damage, and increased coagulation promote clot formation, as in atrial fibrillation, ventricular dysfunction, valvular disease, or infective endocarditis; fragments may then detach and embolize. In immunology and infection research, this topic links coagulation with inflammation, microbial infection, and immune-mediated tissue injury, while clinical evaluation supports risk assessment, anticoagulation, antimicrobial treatment when infection is present, and prevention of stroke or organ damage.

To simulate thromboembolic stroke, emboli prepared from heterologous rat blood were injected into the middle cerebral artery, followed by administration of systemic thrombolysis for recanalization. This version of the model is optimized for use in a multi-laboratory network and supports testing multiple candidate therapeutics.

Chronic Thromboembolic Pulmonary Hypertension and Assessment of Right Ventricular Function in the Piglet

Chronic Thromboembolic Pulmonary Hypertension (CTEPH) and Right Ventricular (RV) dysfunction were induced in piglets by progressive obstruction of the pulmonary arteries. Consequences were remarkably similar to those observed in CTEPH patients. This animal model would be a very useful tool for pathophysiology and therapeutic experiments on CTEPH and RV failure.

Modified Mouse Embryonic Stem Cell based Assay for Quantifying Cardiogenic Induction Efficiency

We describe the use of a mouse ES cell based assay to identify critical time windows for Wnt/β-catenin and BMP signal activation during cardiogenic induction. The method provides a standardized platform that reliably quantifies cardiogenic efficiency, and it is applicable to the study of other cell lineages.
