Salmonella Typhimurium

Salmonella Typhimurium is a Gram-negative bacterial pathogen that causes gastroenteritis and serves as an important model for studying host defense and bacterial infection. After ingestion, it uses a type III secretion system encoded by Salmonella pathogenicity island 1 to invade intestinal epithelial cells, while a second system encoded by pathogenicity island 2 supports survival and replication within host cells, including macrophages. These activities trigger innate immune recognition, inflammatory signaling, and recruitment of immune cells to the intestine. Studying Salmonella Typhimurium helps clarify pathogen invasion, intracellular survival, immune evasion, and the development of vaccines or antimicrobial strategies.

Begin with mice bearing solid tumors implanted subcutaneously in the thigh.Inject the mice via the tail vein with a non-virulent, engineered strain of Salmonella typhimurium. The bacteria circulate in the bloodstream and selectively accumulate in the hypoxic, immunosuppressed tumor microenvironment. They carry a plasmid encoding a therapeutic fusion peptide composed of a cell-penetrating peptide and the mitochondrial target domain of a pro-apoptotic protein, along with phage-derived lysis...

An Allelotyping PCR for Identifying Salmonella enterica serovars Enteritidis, Hadar, Heidelberg, and Typhimurium

2011

We describe a multiplex PCR for the rapid detection of Salmonella enterica serovars Enteritidis, Hadar, Heidelberg, and Typhimurium. Specific Salmonella serovars can be identified by targeting a multiplex PCR to genes and sequences unique to the O-antigen biosynthesis cluster and flagellin of a given serovar. Serovar is assigned then to a Salmonella isolate based on the appearance of specific, size amplicons (PCR product) corresponding to the target allele.
