T Cell Mediated Killing

T cell-mediated killing is an immune process in which cytotoxic T cells identify and eliminate infected, abnormal, or tumor cells, helping control intracellular threats and maintain tissue health. After recognizing a specific peptide presented by major histocompatibility complex class I molecules, the T cell forms an immunological synapse and releases perforin and granzymes, which activate target-cell apoptosis; Fas-Fas ligand signaling can provide an additional death pathway. This mechanism is central to antiviral and antitumor immunity and informs research on immune deficiencies, chronic infections, transplant rejection, and cancer immunotherapies. Understanding its regulation also helps explain how immune responses destroy targets while limiting damage to healthy tissue.

(2017)This video demonstrates the procedure for detecting bacteriocin-mediated killing by spotting supernatant from an antibiotic-induced donor strain onto a soft agar overlay containing a test strain. Clear zones formed after incubation indicate bacterial inhibition.

Many infections elicit a strong CTL response, but occasionally, the quantity of responding cells does not correlate to control of the pathogen1. One measure of CTL quality is their ability to kill specifically2. CFSE labeling of target cells can be used to investigate this CTL response quality in vivo3,4.
