Aggregation-induced Emission

Aggregation-induced emission (AIE) is a photophysical phenomenon in which certain molecules emit stronger fluorescence after clustering, offering useful advantages over conventional dyes that become dim when aggregated. In dilute solution, intramolecular rotations and vibrations dissipate absorbed energy without light emission; aggregation restricts these motions, preserves excited-state energy, and activates bright fluorescence. In medicine, AIE luminogens support sensitive bioimaging, cellular tracking, and detection of biomolecules or disease-related signals. Their resistance to aggregation-caused quenching can improve contrast and signal stability, supporting the development of diagnostic probes, therapeutic monitoring tools, and integrated theranostic platforms.

This video demonstrates turbidimetric analysis to assess collagen-induced platelet aggregation in pre-treated, washed human platelets. Control wells show collagen-induced aggregation, whereas a high concentration of inhibitors blocks Pannexin-1 channels, thereby inhibiting collagen-induced platelet aggregation, evident from unchanged turbidity.

The video demonstrates a fluorescence resonance energy transfer (FRET) flow cytometry assay to detect the seeding activity of protein aggregates isolated from biological samples. Mammalian cells expressing tau reporter proteins are incubated with liposome transduction complexes containing tau seeds. These seeds mediate the aggregation of the reporter proteins, leading to generating a FRET positive signal in the flow cytometer.
