Dithiothreitol

Dithiothreitol (DTT) is a strong reducing agent used to maintain molecules in a chemically reduced state, making it important in biochemical and medical research. Its two thiol groups donate electrons to reduce disulfide bonds, converting them into free sulfhydryl groups and becoming oxidized to a stable disulfide form. In protein analysis, DTT helps unfold disulfide-linked proteins and preserve target proteins during sample preparation, electrophoresis, and laboratory assays. These properties support investigations of protein structure, enzyme activity, redox regulation, and disease-related molecular changes, although DTT is generally used as an experimental reagent rather than a routine therapeutic.

Here we present a step-by-step protocol of the long-length electrostatic repulsion-hydrophilic interaction chromatography-tandem mass spectrometry (LERLIC-MS/MS) method. Here, we present a protocol to enable the capture and identification of disease-specific protein-protein interactions from native cells and tissues using chemical probes and mass spectrometry. The resulting interaction datasets are analyzed through a dedicated web-based platform to reveal dynamic network dysfunctions and pathway alterations linked to disease.
