Leukemic Cell Co-culture

Leukemic cell co-culture is an in vitro model that grows leukemia cells together with other cell types to reproduce cellular interactions within the bone marrow or other disease-relevant environments. In this system, direct contact or soluble factors exchanged through shared media influence leukemic-cell survival, proliferation, differentiation, and response to treatment, while stromal, immune, or endothelial cells can provide supportive signals. By capturing these microenvironmental effects, co-culture models help researchers investigate leukemia biology, identify mechanisms of drug resistance, evaluate candidate therapies, and assess how nonmalignant cells shape disease progression. They therefore complement simplified monoculture experiments and support more physiologically relevant studies in medicine.

2D Co-culture of Leukemic Cells with BMSCs: A Method to Obtain Subpopulations of Leukemic Cells

2023

This video describes the technique for long term 2D co-culture of leukemia cells with bone marrow stromal cells to study their interaction with the bone marrow microenvironment. The 2D coculture model utilizes the acute lymphoblastic leukemia cells and the bone marrow stromal or osteoblasts that influence tumor cell phenotype. This study will support investigation of the mechanisms that underlie bone marrow supported survival of leukemic cells during chemotherapy exposure.

2023

This video describes a method for spatial separation of three leukemic cell subpopulations: the suspended leukemic cells freely floating in the media, phase bright leukemic cells adhered to the surface of the mesenchymal stromal cell monolayer, and the phase dim leukemic cells that have migrated beneath the mesenchymal stromal cell monolayer from 2D co-culture.

2023

This video explains a quantitative reverse phase high-performance liquid chromatography (RP-HPLC) method to measure metabolism of adenosine and associated purines generated by consumption of AMP in extracellular environment of leukemic cancer cells. The protocol describes a sensitive, specific, reliable and reproducible RP-HPLC assay developed and validated for the quantification of extracellular purine nucleotides and nucleosides produced by purified chronic lymphocytic leukemia (CLL) cells...
