Mitochondrial Permeability

Mitochondrial permeability describes how readily substances cross mitochondrial membranes, a property that controls energy production, ion balance, and cell survival. In particular, calcium overload, oxidative stress, or loss of membrane potential can trigger opening of the mitochondrial permeability transition pore, allowing solutes and water to enter, dissipating the proton gradient, and promoting matrix swelling and cytochrome c release. In medicine, altered mitochondrial permeability is linked to ischemia-reperfusion injury, neurodegeneration, and cardiovascular disease, making the permeability transition pore an important target for studying cell death and developing therapies that protect mitochondrial function.

A spectrofluorometric protocol for the measurement of the mitochondrial permeability transition pore opening in isolated mouse heart mitochondria is presented here. The assay involves the simultaneous measurement of mitochondria Ca2+ handling, mitochondrial membrane potential and mitochondrial volume. The procedure for obtaining high-quality and functional heart mitochondria is also described.

Mitochondria can utilize the electrochemical potential across their inner membrane (ΔΨm) to sequester calcium (Ca2+), allowing them to shape cytosolic Ca2+ signaling within the cell. We describe a method for simultaneously measuring mitochondria Ca2+ uptake and ΔΨm in live cells using fluorescent dyes and confocal microscopy.
