Ph Indicator Synaptosomes

A pH indicator is a chemical or fluorescent probe that reports acidity, and in synaptosomes it enables measurement of proton dynamics in isolated nerve terminals. In these preparations, acidic synaptic vesicles are maintained by the vacuolar H+-ATPase, while pH-sensitive fluorescence changes as vesicles acidify, alkalinize, fuse with the plasma membrane, or recycle. Measuring these signals helps researchers investigate synaptic vesicle loading, exocytosis, endocytosis, and neurotransmitter release. This approach links vesicle pH to presynaptic function and provides a practical assay for studying neuronal communication, membrane trafficking, and the effects of pharmacological or genetic manipulation.

A Novel In Vitro Live-imaging Assay of Astrocyte-mediated Phagocytosis Using pH Indicator-conjugated Synaptosomes

This protocol presents an in vitro live-imaging phagocytosis assay to measure the phagocytic capacity of astrocytes. Purified rat astrocytes and microglia are used along with pH indicator-conjugated synaptosomes. This method can detect real-time engulfment and degradation kinetics and provides a suitable screening platform to identify factors modulating astrocyte phagocytosis.
