Phase Plate Conversion

Phase plate conversion is an optical method that transforms phase differences in a wave into measurable intensity differences, making otherwise transparent structures visible in microscopy and other imaging systems. It works by placing a phase plate in the back focal plane to selectively shift the phase of the undeviated, or direct, beam relative to light diffracted by the specimen; their interference converts specimen-induced phase variations into brightness contrast. In phase-contrast microscopy, this approach enables visualization of cells, organelles, and other weakly absorbing samples without staining, supporting live-cell observation and analysis of structure and dynamics.
