Feedback Inhibition

Las reacciones bioquímicas ocurren constantemente en las células, convirtiendo sustancias de partida en diferentes productos, generalmente con la ayuda de enzimas que aceleran las reacciones. Sin enzimas, ¡la mayoría de las reacciones tardarían demasiado en ocurrir para que fueran útiles para la célula! Dado que las enzimas ayudan a controlar la velocidad de las reacciones, su actividad se regula de modo que se mantengan en la célula cantidades apropiadas de materiales de partida, metabolitos intermedios y productos. La acumulación o el agotamiento excesivos de sustancias pueden tener consecuencias desastrosas para la salud de las células y del organismo. En un proceso regulador llamado inhibición por retroalimentación, el producto de una reacción inhibe una enzima en un paso anterior de la ruta metabólica que la produjo. El producto se une a un sitio regulador de la enzima, que es distinto del sitio activo que se une al sustrato, lo que ralentiza o detiene su propia producción. Cuando el producto se une al sitio regulador, provoca un cambio conformacional en la enzima que le impide unirse al sustrato y continuar catalizando la reacción. Esto evita que se acumulen niveles excesivamente altos del producto en la célula. Cuando su nivel desciende lo suficiente, el producto ya no inhibe la enzima y la reacción puede continuar como de costumbre.

Cells use enzymes to run biochemical reactions and make the products they need. These reactions are happening constantly, and enzymes speed them up so they happen fast enough to matter. Enzyme activity also has to be regulated. Cells need the right amounts of starting materials, intermediate metabolites, and final products.

If these substances build up too much or drop too low, the cell can be harmed. That is why cells use control systems to keep metabolic levels balanced. One important control process is feedback inhibition. In this process, the final product of a reaction slows an enzyme from an earlier step in the same pathway.

The product binds to a regulatory site on the enzyme. This site is different from the active site, which is where the substrate normally binds. Binding at the regulatory site changes the enzyme’s shape. That conformational change prevents the enzyme from binding the substrate and keeps the reaction from continuing.

Feedback inhibition helps stop too much product from accumulating in the cell. When product levels fall, the product no longer inhibits the enzyme. The pathway can then start again and continue producing the molecule as needed.
