Membrane Lipids

Tres tipos de lípidos están presentes en la membrana: fosfoglicéridos, esfingolípidos y esteroles.
Los fosfoglicéridos, también conocidos como glicerofosfolípidos, contienen una columna vertebral de glicerol con un grupo fosfato hidrofílico y dos cadenas de acilo graso hidrofóbico, generalmente de 16 a 22 átomos de carbono de largo.
Algunos de los fosfoglicéridos más comunes presentes en la membrana celular son la fosfatidilcolina, la fosfatidiletanolamina, la fosfatidilserina y el fosfatidilinositol, formados por la unión de colina, etanolamina, serina e inositol al grupo fosfato.
Los esfingolípidos son lípidos que contienen una columna vertebral de esfingosina. La esfingosina es un aminoalcohol con una larga cadena de hidrocarburos insaturados. 
El esfingolípido más simple es la ceramida, que se forma cuando una molécula de ácido graso se une al grupo amino de la esfingosina.
El grupo hidroxilo restante de la ceramida puede formar ésteres con otros grupos químicos como la fosfocolina o los carbohidratos, formando esfingomielina o glicolípidos, respectivamente.
El tercer tipo de lípidos de membrana, los esteroles, como el colesterol, no pueden formar de forma independiente una bicapa lipídica. En cambio, se intercalan dentro de las bicapas de fosfolípidos y actúan como un amortiguador de fluidez para la membrana.

Los lípidos son un componente esencial de todas las membranas biológicas. El contenido medio de lípidos en las membranas de los mamíferos es del 50 %, aunque puede ser tan bajo como un 20% en la membrana mitocondrial interna o tan alto como un 80 % en la vaina de mielina presente alrededor de las células nerviosas.

La fosfatidilcolina, la fosfatidiletanolamina, la fosfatidilserina y la esfingomielina son los fosfolípidos más comunes presentes en las membranas de los mamíferos. En pH fisiológico, la fosfatidilserina está cargada negativamente, mientras que las otras tres permanecen neutras. En los lípidos de membrana, el número de átomos de carbono presentes en los ácidos grasos y su grado de saturación pueden variar. Los ácidos grasos pueden ser completamente saturados sin dobles enlaces, monoinsaturados con un solo doble enlace o poliinsaturados con múltiples dobles enlaces.

Los glicolípidos comprenden una clase fisiológicamente importante de lípidos de membrana que siempre están presentes en el lado externo o extracelular de la membrana. En las eucariotas se encuentran tanto en la membrana plasmática como en las membranas intracelulares. El galactocerebrósido, un glicolípido simple, existe en abundancia en las vainas de mielina junto con la esfingomielina. Membrane lipids give biological membranes their structure and help set their properties. In mammalian cells, lipids make up about 50% of the membrane on average. The amount can be much lower in the inner mitochondrial membrane or much higher in the myelin sheath around nerve cells.

Several phospholipids are common in mammalian membranes. These include phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, and sphingomyelin. At physiological pH, phosphatidylserine carries a negative charge, while the other three are neutral.

The fatty acids in membrane lipids can differ in carbon length and in saturation. Some are fully saturated, which means they have no double bonds. Others are monounsaturated with one double bond, or polyunsaturated with multiple double bonds.

Glycolipids are an important membrane lipid class that stays on the outer, or extracellular, side of the membrane. In eukaryotes, they are found in both the plasma membrane and intracellular membranes. Galactocerebroside is a simple glycolipid found in large amounts in myelin sheaths along with sphingomyelin.

Gangliosides are the most complex glycolipids. They are negatively charged because they contain sialic acid, and they can make up as much as 10% of the total lipids in nerve cell membranes. Their roles are also studied in viral and genetic diseases. GM1 acts as a cell surface receptor for cholera toxin, and excess GM2 ganglioside is seen in Tay-Sachs disease.

Sterols are another major class of membrane lipids. Plant-specific sterols can make up 30% to 50% of the total lipids in the plant plasma membrane. In eukaryotic membranes, cholesterol is also common and can account for almost 50% of the total lipids. Cholesterol in human cell membranes is slightly different from ergosterol, which is a common lipid in fungal membranes.
