En la síntesis de ésteres malónicos, los β-diésteres que contienen protones α ácidos en presencia de una base y un haluro de alquilo generan ácidos acéticos sustituidos a través de intermediarios de éster malónico alquilados.
Inicialmente, la base elimina el protón α del diéster para generar un ion enolato nucleofílico que está altamente estabilizado por tres estructuras de resonancia. 
El alquilamiento del carbono aniónico a través de un mecanismo SN2 produce un intermediario de éster malónico de alquilo. Este se somete a una hidrólisis seguida de acidificación para dar el β-diácido. 
El β-diácido, a altas temperaturas, se somete a un proceso concertado que expulsa CO2 para producir un enol. Esto se tautomeriza rápidamente a la forma cetogénica estable del ácido acético sustituido. 
Como la reacción comprende la eliminación de un solo protón, el ácido resultante es monosustituido.

El ꞵ-diácido resultante es inestable a altas temperaturas y elimina fácilmente el CO_2 a través de un estado de transición cíclico de seis miembros, formando un enol. Malonic ester synthesis is a way to make alpha-substituted carboxylic acids from beta-diesters such as diethyl malonate and alkyl halides. It builds a new carbon-carbon bond and then turns the product into a carboxylic acid.

The reaction starts by removing the acidic alpha hydrogen from the beta-diester. This forms a doubly stabilized enolate ion, which is a nucleophile, meaning it can attack an electron-poor partner.

The enolate reacts with the alkyl halide by an SN2 mechanism. That step gives an alkylated malonic ester intermediate with the new carbon-carbon bond in place.

Next, aqueous acid or base hydrolyzes both ester groups. This forms a 1,3-dicarboxylic acid. When heated, that diacid is unstable and loses CO2 through a cyclic six-membered transition state.

The decarboxylation produces an enol, and the enol then tautomerizes to the more stable keto form. The final product is a monosubstituted carboxylic acid. If the deprotonation and alkylation steps are repeated before hydrolysis and decarboxylation, a disubstituted carboxylic acid can be made instead.
