Forces Acting on Chromosomes

Durante la mitosis, los cromosomas biorientados oscilan para estabilizar sus uniones de huso y posteriormente se ensamblan a lo largo de la placa de metafase en el ecuador del huso mitótico.
Múltiples fuerzas actúan sobre los cromosomas después de que se unen al huso mitótico.
Una fuerza importante hacia los polos ejercida a lo largo del microtúbulo del cinetocoro, como resultado de la despolimerización del extremo positivo, tira del cinetocoro y su cromosoma asociado hacia el polo del huso. En el cinetocoro, los complejos proteicos Ndc80 unen el cinetocoro al microtúbulo a través de múltiples uniones de baja afinidad a lo largo de los lados de los microtúbulos.

Durante la mitosis, los movimientos cromosómicos se producen mediante la interacción de múltiples fuerzas a nivel de piconewton. En la prometafase, estas fuerzas ayudan en el ensamblaje o congreso de los cromosomas en el plano ecuatorial, lo que eventualmente conduce a su alineación en la placa metafásica. Las fuerzas que actúan sobre los cromosomas dependen del espacio y del tiempo; por lo tanto, varían con la posición de los cromosomas a medida que la célula avanza a través de la mitosis.

Los microtúbulos y las proteínas motoras ejercen dos tipos de fuerzas sobre los cromosomas: hacia los polos y hacia los polos, también conocidas como fuerzas de eyección polar. La despolimerización de los microtúbulos del cinetocoro genera la fuerza hacia el polo y tira del cromosoma hacia el polo del huso. La acción simultánea pero desigual de las fuerzas hacia los polos y de expulsión polar provoca la oscilación de los cromosomas durante la prometafase; sin embargo, durante la metafase, las cromátidas hermanas biorientadas experimentan fuerzas iguales pero opuestas. Esto crea suficiente tensión para silenciar la vía del punto de control del conjunto del huso y permite que las células entren en anafase. En la anafase, las fuerzas hacia los polos actúan sobre las cromátidas hermanas, lo que da como resultado su segregación exitosa a las células hijas.
