The Central Dogma

El dogma central de la biología molecular establece que la información codificada en el ADN se transfiere al ARN, que luego dirige la síntesis de proteínas en función de estas instrucciones.
En primer lugar, durante la transcripción, el ADN se utiliza como molde para sintetizar ARN mensajero o ARNm.
El ARNm representa una copia de la hebra codificante del ADN, excepto que las timinas son reemplazadas por uracilos.
A continuación, el ARNm se traduce en una cadena de aminoácidos por un ribosoma.
Aquí, grupos de tres nucleótidos en el ARNm, también conocidos como codones, se unen a secuencias complementarias o anticodones en las moléculas de ARN de transferencia o ARNt.
Cada ARNt se une a un aminoácido en particular, dependiendo del codón específico.
Por ejemplo, el codón CCA se une a un ARNt unido a la prolina, mientras que el AGC se une a un ARNt unido a la serina.
De esta manera, el código genético especifica el orden de los aminoácidos en el polipéptido resultante, que se procesa posteriormente para convertirse en una proteína funcional.

El ARN es el eslabón perdido entre el ADN y las proteínas

A principios del siglo XX, los científicos descubrieron que el ADN almacena toda la información necesaria para las funciones celulares y que las proteínas realizan la mayoría de estas funciones. Sin embargo, los mecanismos para convertir la información genética en proteínas funcionales permanecieron desconocidos durante muchos años. Inicialmente se creía que un solo gen se convertía directamente en su proteína codificada. Dos descubrimientos cruciales en células eucariotas cuestionaron esta teoría. Primero, la producción de proteínas no ocurre en el núcleo. En segundo lugar, el ADN no está presente fuera del núcleo. Estos hallazgos provocaron la búsqueda de una molécula intermediaria que conecte el ADN con la producción de proteínas. Se descubrió que esta molécula intermediaria era ARN.

El ARN se sintetiza en el núcleo utilizando el ADN como plantilla. El ARN recién sintetizado tiene una secuencia similar a la cadena de ADN, excepto que la timina es reemplazada por uracilo. En eucariotas, este transcrito primario se procesa aún más, eliminando las regiones no codificantes de la proteína, tapando el extremo 5' y agregando una cola poli-A 3', para crear ARNm maduro, que entonces se exporta al citoplasma.

El código genético es redundante

Las proteínas se crean a partir de 20 aminoácidos en las eucariotas. La combinación de cuatro nucleótidos en conjuntos de tres proporciona 64 (43) codones posibles. Esto significa que más de un codón puede codificar un aminoácido individual. Por tanto, se dice que el código genético es redundante o degenerado. A menudo, pero no siempre, los codones que especifican los mismos aminoácidos difieren sólo en el tercer nucleótido del triplete. Por ejemplo, los codones GUU, GUC, GUA y GUG representan el aminoácido valina. Sin embargo, AUG es el único codón que representa el aminoácido metionina. El codón AUG es también el codón donde comienza la síntesis de proteínas y, por lo tanto, se denomina codón de inicio. La redundancia en el sistema minimiza los efectos nocivos de las mutaciones. Por ejemplo, una alteración en la tercera posición del codón podría no cambiar necesariamente el aminoácido codificado, por lo que no provocaría ningún cambio en la funcionalidad de la proteína.

El código genético es universal

Con algunas excepciones, la mayoría de los organismos procarióticos y eucariotas utilizan el mismo código genético para la síntesis de proteínas. Esta universalidad del código genético ha permitido avances en la investigación científica en el campo de la agricultura y la medicina. Por ejemplo, ahora la insulina humana se puede fabricar a gran escala en bacterias utilizando tecnología de ADN recombinante. La tecnología del ADN recombinante implica el uso de material genético de diferentes especies.
