Transcription

La transcripción es el proceso de sintetizar ARN a partir de una plantilla de ADN.
En el ADN molde, el complejo de preiniciación de la transcripción se ensambla alrededor del promotor central de un gen. El promotor incluye la caja TATA y una secuencia de iniciador. Los factores de transcripción generales se unen en la caja TATA, mientras que la secuencia iniciadora contiene el sitio de inicio de la transcripción.
Una vez que se unen los componentes necesarios, el complejo de preiniciación desenrolla un corto tramo del ADN aguas arriba del sitio de inicio de la transcripción. Después de eso, los factores de transcripción generales se disocian de la hebra y la ARN polimerasa comienza a producir una nueva hebra de ARNm.
Los nucleótidos se agregan uno por uno, y la síntesis del ARNm ocurre en una dirección de cinco primos a tres primos, leyendo de la hebra moltal, excepto que las timidinas son reemplazadas por uridinas.
Esta hebra de ARNm recién creada representa una copia de la información en la hebra codificante. La síntesis continuará hasta que se encuentre una secuencia de terminación, que liberará el ARNm recién fabricado.

La transcripción es la síntesis de ARN a partir de una secuencia de ADN mediante la ARN polimerasa. Es el primer paso en la producción de una proteína a partir de una secuencia genética. Además, muchas otras proteínas y secuencias reguladoras participan en la correcta síntesis del ARN mensajero (ARNm). La regulación transcripcional es responsable de la diferenciación de diferentes tipos de células y, a menudo, de la respuesta celular adecuada a las señales ambientales.

La transcripción puede producir diferentes tipos de moléculas de ARN

En las eucariotas, el ADN se transcribe primero en pre-ARNm, que puede procesarse posteriormente en un ARNm maduro que sirva como plantilla para la síntesis de proteínas. Sin embargo, en los procariotas, la traducción del ARN a polipéptidos puede comenzar mientras la transcripción aún está en curso. La transcripción también puede producir diferentes tipos de moléculas de ARN que no codifican proteínas, como microARN (miARN), ARN de transferencia (ARNt) y ARN ribosómico (ARNr), todos los cuales afectan la síntesis de proteínas.

La regulación de la transcripción es fundamental para el desarrollo

Con pocas excepciones, todas las células del cuerpo humano tienen la misma información genética, pero diferentes tipos de células regulan de manera diferencial la transcripción durante el desarrollo. Específicamente, la regulación transcripcional desempeña un papel central en la diferenciación celular: el proceso de producción de células especializadas, como las células musculares, a partir de células precursoras menos especializadas. Algunos genes de las células precursoras deben activarse y otros desactivarse para producir células especializadas.

Este proceso de diferenciación celular está orquestado por proteínas de unión al ADN llamadas factores de transcripción que controlan el nivel de transcripción de los genes. Por ejemplo, durante el desarrollo temprano de los vertebrados, las células de la capa de ectodermo del embrión en desarrollo reciben varias señales de inducción de proteínas como BMP, WNT y SHH.
