La preparación de la muestra es un paso crítico para obtener resultados de análisis precisos. Comprende tres objetivos principales: extracción, purificación y concentración.
En primer lugar, la extracción consiste en aislar una mezcla con el analito de interés de una matriz de muestra. En segundo lugar, si hay sustancias interferentes, se eliminan para obtener una muestra pura. En tercer lugar, si el analito extraído es de baja concentración, se concentra a un nivel que se puede detectar con precisión.
Las formas comunes de la muestra son sólido, líquido y gaseoso. Los trozos sólidos demasiado grandes para ser analizados directamente se pulverizan mediante métodos físicos como la molienda, la trituración y la molienda.
El tamizado y la mezcla de esta mezcla dan como resultado un polvo fino homogéneo, que luego se seca y se disuelve en un solvente adecuado para extraer el analito en la solución.

Después de la molienda, la muestra se tamiza y se mezcla para garantizar una composición uniforme. Luego, la muestra en polvo se disuelve en un disolvente adecuado para extraer el analito de la matriz sólida. La elección del disolvente depende de la naturaleza del analito y de las características de solubilidad de los componentes de la muestra.

Las muestras líquidas que contienen partículas sólidas suspendidas o precipitados se someten convencionalmente a filtración o centrifugación para separar las impurezas sólidas del líquido. La filtración implica pasar el líquido a través de un filtro, mientras que la centrifugación utiliza la fuerza centrífuga (de giro) para separar los sólidos de la fase líquida. Sample preparation depends on the state and form of the sample before analysis. It helps make the sample ready for accurate measurement. The main goal is to extract the analyte, which is the substance being measured, and remove parts that could interfere with the analysis.

Solid samples often need the most handling. Large solids are usually ground, crushed, or milled to make them smaller and increase surface area. The sample is then sieved and mixed so it has a uniform composition. After that, the powdered material is dissolved in a suitable solvent to pull the analyte out of the solid matrix. The best solvent depends on the analyte and on how well the sample components dissolve.

Liquid samples may also need treatment before analysis. If they contain suspended solids or precipitates, they are usually filtered or centrifuged to remove the unwanted particles. Filtration works by passing the liquid through a filter. Centrifugation uses spinning force to separate the solid material from the liquid phase.

When the analyte is present in trace amounts, liquid samples may need a concentration step. This increases the analyte level so it is easier to detect. Sample preparation is usually not needed for gaseous analytes because the gas sample can be introduced directly into the analytical instrument.
