Enzyme-coupled Assay

Enzyme-coupled assays are analytical methods that measure a biochemical reaction indirectly by linking it to a second, readily detectable enzyme reaction. The target reaction produces or consumes a substrate that the coupling enzyme transforms into a colored, fluorescent, or otherwise measurable product, allowing signal intensity to reflect reaction progress under defined conditions. In biochemistry, these assays help quantify metabolites, enzyme activities, and reaction kinetics, particularly when the primary reaction lacks a convenient detection signal. They support substrate analysis, inhibitor screening, clinical measurements, and mechanistic studies by converting otherwise difficult-to-observe molecular changes into reproducible experimental readouts.

Enzyme-Linked Immunosorbent Assay to Verify Chemically-Coupled Antibody-Antigen Conjugates

2025

This video demonstrates the detection of chemically conjugated antibody-antigen complexes via the sandwich enzyme-linked immunosorbent assay, or ELISA, technique. The captured antibody binds to the specific epitope of the antigen of the antibody-antigen conjugate, which can be later visualized using an enzyme-linked secondary antibody.

Enzyme Assays and Kinetics

2023

Enzyme kinetics describes the catalytic effects of enzymes, which are biomolecules that facilitate chemical reactions necessary for living organisms. Enzymes act on molecules, referred to as substrates, to form products. Enzyme kinetic parameters are determined via assays that directly or indirectly measure changes in substrate or product concentration over time. This video will cover the basic principles of enzyme kinetics (including rate equations) and kinetic models. The concepts governing...

SAMHD1 is a deoxynucleoside triphosphate triphosphohydrolase with critical roles in human health and disease. Here we present a versatile enzyme-coupled SAMHD1 activity assay, deployed in a 384-well microplate format, that allows for the evaluation of small molecules and nucleotide analogues as SAMHD1 substrates, activators, and inhibitors.

Insoluble Chromogenic Biomass-Based Enzyme Screening: An Assay to Evaluate the Degradation Activity of Plant Biomass-Degrading Enzymes

2025

This video demonstrates an assay to screen enzymes effective in degrading complex polysaccharides in plant biomass using insoluble chromogenic biomass or ICB. The enzymes hydrolyze ICB to release soluble dye-linked oligosaccharides, and the enzyme activity is assessed by colorimetric analysis of the reaction product.

Chromogenic Polymer Hydrogel-Based Enzyme Screening Assay: A High Throughput Method to Screen the Carbohydrate-Active Enzymes Using Synthetic Substrates

2025

In this video, we demonstrate a high throughput assay for carbohydrate-active enzyme screening using insoluble chromogenic polymer hydrogel substrates. The enzyme degrades these substrates into a soluble colored product that can be quantified by measuring the color intensity of the product.
