Inclusion Body Solubilization

Inclusion body solubilization is a biochemical method for dissolving dense, insoluble aggregates of recombinant proteins that accumulate in microbial cells, making the target protein available for purification and refolding. The process typically uses strong denaturants such as urea or guanidine hydrochloride, often with reducing agents, to disrupt hydrophobic interactions, hydrogen bonding, and incorrect disulfide bonds that stabilize the aggregates. After solubilization, controlled removal or dilution of the denaturant can promote protein refolding, although conditions must be optimized to limit aggregation and preserve activity. This approach supports the production of enzymes, antibodies, and other valuable proteins for structural studies, diagnostics, and biotechnology.
