Side-chain Protecting Groups

Side-chain protecting groups are temporary chemical modifications that mask reactive amino acid side chains during peptide synthesis, preventing unwanted reactions while the peptide backbone is assembled. They work by converting functional groups such as amines, carboxylic acids, alcohols, and thiols into less reactive derivatives that remain stable under selected coupling conditions and can later be removed selectively. In biochemistry, this strategy enables controlled formation of peptide bonds, supports orthogonal protection schemes, and improves product purity by limiting side reactions. Careful selection of protecting groups and deprotection conditions is essential for synthesizing peptides, modified proteins, and other biologically active molecules.
