Bacterial Enrichment

Bacterial enrichment is a microbiological process that increases the relative abundance of selected bacteria in a mixed sample, making them easier to detect, isolate, and study. It works by placing the sample in an enrichment medium or environment with conditions that favor the target organisms, such as specific nutrients, temperature, pH, oxygen availability, or selective inhibitors, while limiting competing microbes. In biology, bacterial enrichment supports the recovery of low-abundance species from environmental, clinical, food, and industrial samples. The resulting cultures can be used for identification, physiological analysis, molecular testing, and investigation of microbial functions or interactions.

Source: Armbruster, K. M., Meredith, T. C. Enrichment of Bacterial Lipoproteins and Preparation of N-terminal Lipopeptides for Structural Determination by Mass Spectrometry. J. Vis. Exp. (2018)This video demonstrates the enrichment of bacterial lipoproteins using a non-ionic detergent that undergoes phase separation upon warming. Through sequential temperature shifts and centrifugation steps, lipoproteins are selectively partitioned into the detergent phase, while non-lipoprotein contaminants...

Here, we present a protocol to enable whole-genome sequencing of bacterial sexually transmitted infections from clinical samples using target enrichment. This novel, syndromic panel-based method overcomes challenges of abundant human DNA and low bacterial loads, facilitating genomic surveillance for Chlamydia trachomatis, Neisseria gonorrhoeae, Treponema pallidum, and Mycoplasma genitalium.

2026

All animal procedures described here must be conducted in accordance with institutional animal ethics guidelines and approved by IACUC. All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Environmental enrichment involves objects and practices that encourage natural behaviors, improve psychological well-being, and promote better welfare in laboratory rodents. Environmental enrichment should be tailored to the...

Enrichment of Anode-Respiring Bacteria Using an On-Site Electrochemical System

Source: Okamoto, A., et al. Self-standing Electrochemical Set-up to Enrich Anode-respiring Bacteria On-site. J. Vis. Exp. (2018)This video demonstrates the use of an on-site electrochemical system to enrich anode-respiring bacteria (ARB) from natural reservoirs. A redox gradient is established by placing the anode in oxygen-free deep water and the cathode in oxygen-rich surface water. ARB form biofilms and transfer electrons to the anode, which flows to the cathode, where oxygen is reduced,...

Bacterial Infection Quantitation Assay: A Method to Quantify Intraocular Bacterial Load in Mouse Model of Bacterial Endophthalmitis

2025

This video describes the technique of quantifying bacterial infection from eye homogenates obtained from a murine model of bacterial endophthalmitis caused by Bacillus cereus. This technique helps study the host immune response and its dependency on the host and ocular bacterial gene expression in murine models.
