Gene Specific Primers

Gene-specific primers are short synthetic DNA oligonucleotides designed to bind selected sequences within or near a target gene, enabling focused analysis of that gene. During polymerase chain reaction (PCR), forward and reverse primers anneal to complementary strands at defined temperatures and provide starting points for DNA polymerase, which extends them to produce millions of copies of the target region. Primer sequence, length, and annealing conditions influence amplification specificity and efficiency. In biology, gene-specific primers support genotyping, mutation detection, cloning, gene expression analysis through reverse transcription PCR, and sequencing, helping researchers investigate genetic variation and gene function.

Most common gut content analyses of macroinvertebrates are visual. Requiring intense knowledge about morphological diversity of prey organisms, they miss soft bodied prey and, due to strong comminution of prey, are nearly impossible for some organisms, including amphipods. We provide detailed, novel genetic approaches for macroinvertebrate prey identification in the diet of amphipods.

An Allele-specific Gene Expression Assay to Test the Functional Basis of Genetic Associations

Genetic associations often remain unexplained at a functional level. This method aims to assess the effect of phenotype-associated genetic markers on gene expression by analyzing cells heterozygous for transcribed SNPs. The technology allows accurate measurement by MALDI-TOF mass spectrometry to quantify allele-specific primer extension products.

Primer-Free Aptamer Selection Using A Random DNA Library

SELEX protocols comprise multiple rounds of selection, each of which require regeneration of bound ligands, which in turn require fixed primer sequences flanking the random library regions. These fixed primer sequences can interfere with the selection process (false positives and negatives). Here we present a primer-free protocol.
