Inbreeding

Inbreeding is the reproduction of individuals that share a recent common ancestor, increasing genetic similarity within offspring and populations. Because related parents are more likely to carry the same alleles, inbreeding raises homozygosity and can expose harmful recessive variants, sometimes causing inbreeding depression such as reduced fertility, survival, or growth. Biology uses inbreeding to study inheritance, genetic variation, and the effects of natural or artificial selection, while conservation programs monitor it to preserve population fitness and minimize the loss of genetic diversity. Controlled inbreeding can also establish stable laboratory strains or enhance desired traits in domesticated organisms.

In this protocol, methods for conducting inbreeding crosses, and for evaluating the success of those crosses, are described for the ant Vollenhovia emeryi. These protocols are important for experiments aimed at understanding the genetic basis of sex determination systems in Hymenoptera.

Maintaining Aedes aegypti Mosquitoes Infected with Wolbachia

Aedes aegypti mosquitoes infected with Wolbachia are being released into natural populations to suppress the transmission of arboviruses. We describe methods to rear Ae. aegypti with Wolbachia infections in the laboratory for experiments and field release, taking precautions to minimize laboratory adaptation and selection.

Source: Kay Stewart, RVT, RLATG, CMAR; Valerie A. Schroeder, RVT, RLATG. University of Notre Dame, IN Millions of mice and rats are bred for use in biomedical research each year. Worldwide, there are several large commercial breeding facilities that supply mice to research laboratories, but many facilities also choose to breed mice and rats in-house to reduce costs and increase research options. When breeding in the animal facility, researchers are able to manipulate the genetics of the...
