Braf Pten Mouse

Braf Pten mouse models are genetically engineered mice used to study how oncogenic BRAF signaling and loss of the tumor suppressor PTEN promote cancer development, particularly melanoma. Typically, a conditional Braf mutation such as BRAF V600E is activated in melanocytes while floxed Pten alleles are deleted through tissue-specific or inducible Cre recombinase, combining MAPK pathway activation with impaired PI3K-AKT regulation. This cooperation drives tumor initiation and progression in a defined, immunocompetent setting. Researchers use these mice to investigate tumor biology, treatment resistance, immune responses, and therapeutic strategies targeting BRAF, MEK, PI3K, or related pathways.

Melanoma is a very aggressive disease that quickly spreads to other organs. This protocol describes the application of ultra-high-frequency ultrasound imaging, coupled with 3D rendering, to monitor the volume of the inguinal lymph nodes in the Braf/Pten mouse model of metastatic melanoma.

The goal of this video is to demonstrate how to perform automated DNA extraction from formalin-fixed paraffin-embedded (FFPE) reference standard cell lines and digital droplet PCR (ddPCR) analysis to detect rare mutations in a clinical setting. Detecting mutations in FFPE samples demonstrates the clinical utility of ddPCR in FFPE samples.

Mice are widely used to study gestational biology. However, pregnancy termination is required for such studies which precludes longitudinal investigations and necessitates the use of large numbers of animals. Therefore, we describe a non-invasive technique of high-frequency ultrasonography for early detection and monitoring of post-implantation events in the pregnant mouse.
