Brain Tumor Tissue Sections

Brain tumor tissue sections are thin, prepared slices of tumor and surrounding brain tissue used to examine cancer at the cellular and tissue levels. Researchers preserve, embed, and cut specimens before applying stains or molecular labels that reveal cellular architecture, tumor boundaries, vascular features, and biomarker distribution under microscopy. In cancer research, these sections support histopathological classification, assessment of tumor heterogeneity, comparison of malignant and nonmalignant regions, and evaluation of treatment-related changes. When paired with immunohistochemistry, fluorescence imaging, or spatially resolved molecular methods, brain tumor tissue sections connect microscopic structure with disease mechanisms and can inform diagnosis, therapeutic studies, and the development of more precise models.

Source: Núñez, F. J., et al. Evaluation of Biomarkers in Glioma by Immunohistochemistry on Paraffin-Embedded 3D Glioma Neurosphere Cultures. J. Vis. Exp. (2019).This video demonstrates an immunohistochemical technique for labeling tumor biomarkers in paraffin-embedded brain tumor neurospheres. The neurosphere sections undergo paraffin removal and rehydration, followed by treatment to expose biomarker sites. Primary and secondary antibodies are then applied to label these biomarkers. The...

Source: Maiti, P. et. al.,. Labeling and Imaging of Amyloid Plaques in Brain Tissue Using the Natural Polyphenol Curcumin. J. Vis. Exp. (2019)This video demonstrates the imaging procedure of amyloid-beta plaques in brain tissue sections using immunolabeling and curcumin staining. Immunolabeling provides specificity for amyloid-beta, while curcumin selectively binds β-sheet-rich fibrils, enhancing visualization. The co-localization of red and green fluorescence confirms the presence of plaques.
