Cell Isolation Method

Cell isolation methods are laboratory techniques used to separate individual cells or defined cell populations from tissues, blood, tumors, or mixed cultures for controlled analysis. They typically combine mechanical or enzymatic dissociation with filtration, centrifugation, density-based separation, or affinity-based selection, using differences in cell size, density, surface markers, or viability to enrich target populations while preserving cellular function. In cancer research, isolated tumor cells, immune cells, and stromal populations support flow cytometry, genomic and transcriptomic profiling, drug-response testing, and organoid development. Reliable isolation improves the resolution of tumor heterogeneity studies and helps connect cellular characteristics with disease progression and treatment response.

Here we describe a high efficiency, high yield method for isolation of all of the currently known major subsets of mouse splenic dendritic cells.
