Drug Content Measurement

Drug content measurement is the analytical determination of how much active pharmaceutical ingredient is present in a drug substance or formulation, supporting accurate dosing and quality control. In chemistry, analysts prepare a representative sample, separate or detect its components using methods such as high-performance liquid chromatography, ultraviolet-visible spectroscopy, or mass spectrometry, and compare the signal with calibrated reference standards to calculate concentration. The results help assess potency, content uniformity, stability, and compliance with product specifications. Reliable measurement is essential during pharmaceutical development, manufacturing, release testing, and stability studies because it can reveal degradation, formulation variability, or inaccurate labeling.

A simple and non-destructive technique that measures the average content of drug substances in formulated drug products containing fluorine using low-field fluorine-19 (19F) time-domain (TD) nuclear magnetic resonance (NMR) is presented here. Here we describe a novel high-content chemically induced inflammation assay aiming at the identification of immune-modulatory bioactives. We have successfully combined automated microscopy with custom developed software scripts enabling automated quantification of the inflammatory response as well as further data processing, analysis, mining, and storage.
