Spectrophotometer Readings

Spectrophotometer readings are quantitative measurements of how much light a sample absorbs or transmits at selected wavelengths, helping chemists characterize substances and determine their concentrations. A spectrophotometer passes light through a sample, compares the transmitted intensity with a reference, and reports absorbance or transmittance; under suitable conditions, absorbance follows the Beer-Lambert law and increases with concentration and optical path length. In chemistry, these readings support concentration calculations, reaction-rate monitoring, purity assessment, and identification of compounds based on their absorption spectra. Accurate results depend on appropriate wavelength selection, calibration with blanks or standards, and consistent sample handling.

2023

The spectrophotometer is a routinely used instrument in scientific research. Spectrophotometry is the quantitative measurement of how much a chemical substance absorbs light by passing a beam of light through the sample using a spectrophotometer. In this video, basic concepts in spectrophotometry, including transmittance, absorbance and the Beer-Lambert Law are reviewed in addition to the components of the spectrophotometer. These concepts provide a foundation for how to determine the...

Research articles are structured documents designed to communicate scientific findings efficiently and transparently. Rather than reading them sequentially like a narrative, readers can approach them strategically by understanding their standardized organization. Evaluating the title, authors, and institutional affiliations provides an initial indication of relevance, expertise, and credibility. The Abstract offers a concise summary of the study’s objective, methodology, and main findings,...

Uncertainty in Measurement: Reading Instruments

Counting is the type of measurement that is free from uncertainty, provided the number of objects being counted does not change during the process. Such measurements result in exact numbers. By counting the eggs in a carton, for instance, one can determine exactly how many eggs are there in the carton. Similarly, the numbers of defined quantities are also exact. For example, 1 foot is exactly 12 inches, 1 inch is exactly 2.54 centimeters, and 1 gram is exactly 0.001 kilograms. Quantities...

This communication presents data on the accuracy and reproducibility of the BioSpec-nano UV-VIS spectrophotometer for dsDNA and protein quantitation. Even with ultra-small volumes (1 to 2 L), reproducibility is excellent, while the automated wiping function improves throughput and results in minimal carryover for more precise results.
