Sterile Vegetable Extract

Sterile vegetable extract is a microorganism-free preparation made from vegetable material, providing soluble nutrients and plant-derived compounds for controlled laboratory studies. It is produced by extracting water-soluble components and applying a sterilization approach, such as membrane filtration or heat treatment, under conditions that limit contamination while preserving as much of the extract’s chemical composition as possible. In environmental research, this material can serve as a defined or supplementary nutrient source for microbial enrichment, biodegradation studies, and investigations of plant-derived organic matter. Its sterility improves experimental reproducibility by separating the effects of the extract from uncontrolled microbial growth.

Burch in their book The Principles of Humane Experimental Technique. The 3 R's are replacement, reduction, and refinement of the use of animals in research. The use of cell lines and tissue cultures that originated from research animals is a replacement technique, as it allows for many experiments to be conducted in vitro. Harvesting...

Source: Wunderlin, T., et al. Physical Isolation of Endospores from Environmental Samples by Targeted Lysis of Vegetative Cells. J. Vis. Exp. (2016)This video demonstrates a method to isolate endospores from an environmental sample containing both vegetative cells and various bacteria. The procedure involves filtration, mechanical disruption, and selective chemical treatment to weaken and lyse vegetative cells while preserving endospores. The final step isolates the resilient endospores through...

Source: Laboratory of Jeff Salacup - University of Massachusetts Amherst The distribution of a group of organic biomarkers called glycerol-dialkyl glycerol-tetraethers (GDGTs), produced by a suite of archaea and bacteria, were found in modern sediments to change in a predictable manner in response to air or water temperature1,2. Therefore, the distribution of these biomarkers in a sequence of sediments of known age can be used to reconstruct the evolution of air and/or water temperature on...

Cleaning, Sterilization, and Disinfection

Cleaning, disinfection, and sterilization are the methods that help to break the infection chain and prevent disease. Cleaning The cleaning process usually involves using water with detergents or enzymatic cleaner and removing foreign material from objects and surfaces, including organic material such as body fluids or inorganic material like soil. Cleaning is performed before high-level disinfection and sterilization because foreign materials on the cover of the devices interfere with process...
