Tn5 Transposition

Tn5 transposition is the movement of DNA segments by the Tn5 transposase, a process that reshapes genomes and provides a versatile tool for genetic analysis. The transposase recognizes specific mosaic end sequences, excises the associated DNA, and inserts it at a new genomic location, while engineered Tn5 complexes can simultaneously fragment DNA and add sequencing adapters through tagmentation. In genetics, these properties support insertional mutagenesis, genome-wide mapping of accessible chromatin with ATAC-seq, and rapid preparation of sequencing libraries. Tn5-based methods help researchers investigate gene function, regulatory architecture, and genome organization across diverse biological systems.
