Cfu Lung Analysis

CFU lung analysis is a microbiological method for quantifying viable bacteria in lung tissue or respiratory samples, providing a direct measure of pulmonary infection burden. In this assay, samples are homogenized, serially diluted, and spread on appropriate agar; each viable bacterial cell or clump can produce a visible colony, allowing results to be reported as colony-forming units (CFU), often normalized to tissue mass or sample volume. In immunology and infection research, CFU lung analysis helps assess pathogen growth, clearance by host immune responses, disease severity, and the effects of antimicrobial treatments or vaccines. It remains a practical endpoint for comparing infection outcomes in experimental models.

We illustrate non-surgical delivery of test materials into the lungs of anesthetized mice via the trachea. This method permits lung exposure to bacterial and viral pathogens, cytokines, antibodies, beads, chemicals, or dyes. We further describe harvesting and processing of lungs and lung draining lymph nodes (LDLNs) for flow cytometry.

Source: Yuan, Z., et al. Bronchoalveolar Lavage Exosomes in Lipopolysaccharide-induced Septic Lung Injury. J. Vis. Exp. (2018)This video demonstrates an in vivo mouse model for inducing and analyzing acute lung injury (ALI) using intraperitoneal administration of bacterial lipopolysaccharide. The inflammatory cascade triggered by alveolar macrophages and neutrophils leads to epithelial and endothelial barrier disruption, allowing fluid and immune cells to accumulate in the alveoli. Alveolar...

This paper describes a striaghforward and efficient method of intubating mice for pulmonary function measurements or pulmonary instillation, that allows the mice to recover and be studied at later times. The procedure involves an inexpensive fiberoptic light source that directly illuminates the trachea.
