Cytoarchitecture Preservation

Cytoarchitecture preservation is the maintenance of a tissue’s cellular organization, spatial relationships, and structural features during collection, fixation, processing, and analysis. In neuroscience, careful fixation stabilizes cells and extracellular structures, while controlled sectioning and staining retain features such as cortical layers, neuronal distribution, and connections between anatomical regions. Preserving this organization allows researchers to distinguish healthy from diseased tissue, map brain regions, and relate cellular structure to neural function. It also supports accurate interpretation of histology, immunohistochemistry, and microscopy, making structural integrity essential for studying development, injury, neurodegeneration, and other changes in the nervous system.

Vibratome Sectioning for Enhanced Preservation of the Cytoarchitecture of the Mammalian Organ of Corti

A simple procedure of vibratome sectioning the organ of Corti, followed by immunohistochemistry and confocal microscopy is described. This procedure allows for improved preservation of the fine cytoarchitecture of the mammalian organ of Corti, and consequently allows for accurate quantification of cell types.
