Malachite Green Assay

The Malachite Green Assay is a colorimetric method for detecting inorganic phosphate released during biochemical reactions, making it useful for measuring phosphate-dependent enzyme activity. Under acidic conditions, phosphate reacts with molybdate to form a phosphomolybdate complex, which binds malachite green and produces a green color whose absorbance increases with phosphate concentration. In neuroscience research, this assay can quantify activities such as ATPases and phosphatases in purified proteins, cell extracts, or membrane preparations. Its sensitivity and compatibility with microplate formats support enzyme characterization, screening of modulators, and investigation of molecular processes that regulate neuronal function.

This publication presents a protocol showing how to measure the activity of recombinant protein phosphatase 2A catalytic subunit (PP2Ac) in response to recombinant α-synuclein, β-synuclein, or γ-synuclein proteins using a simple colorimetric assay.
