Plethysmography Recording

Plethysmography recording is a noninvasive method for tracking changes in the volume of a body region or organ, making it useful for measuring physiological signals relevant to neuroscience. Depending on the sensor and site, the method detects volume-related changes caused by blood flow, respiration, or other tissue expansion, often converting them into continuous electrical or optical signals. Researchers use these recordings to quantify peripheral vascular responses, breathing patterns, and autonomic nervous system activity during sensory, cognitive, or emotional tasks. When synchronized with neural or behavioral measures, plethysmography helps relate brain activity to changes in whole-body physiology.

The assessment of respiratory physiology has traditionally relied upon techniques, which require restraint or sedation of the animal. Unrestrained whole-body plethysmography, however, provides precise, non-invasive, quantitative analysis of respiratory physiology in animal models. In addition, the technique allows repeated respiratory assessment of mice allowing for longitudinal studies.

Unrestrained barometric plethysmography is used to quantify the pattern of breathing in awake mice. We show that 15 s segments under a standardized protocol display similar values to an extended duration of quiet breathing. This methodology also allows for the quantification of apnea and augmented breaths during the first hour in the chamber.
