Propidium Iodide Staining

Propidium iodide staining is a fluorescent method for identifying cells with compromised plasma membranes, making it useful for assessing cell viability and death. Because propidium iodide cannot cross intact membranes, it enters damaged or dying cells and intercalates with nucleic acids, producing red fluorescence that can be measured by microscopy or flow cytometry. In neuroscience, researchers use this assay to detect neuronal injury, monitor neurotoxic effects, and quantify cell loss in cultures and tissue samples. Combined with markers such as Annexin V, it can help distinguish membrane disruption associated with late apoptosis or necrosis from viable cells.
