Vesicle Motility

Vesicle motility is the directed movement of membrane-bound compartments within cells, a process that organizes intracellular transport and is essential for neuronal communication. In neurons, motor proteins such as kinesin and dynein move vesicles along microtubules, while actin-based motors and cytoskeletal networks support delivery near synapses; motor activity is regulated by adaptor proteins, cargo identity, and cellular signals. Studying this movement helps explain how synaptic vesicles reach release sites, how neurotransmitter secretion is maintained, and how transport defects can disrupt neuronal function. Experimental analyses of vesicle motility therefore inform research on synaptic physiology, axonal transport, and neurological disease.

Visualization of Vesicle Motilities in Neurons Using Fluorescence Imaging

This video demonstrates how neuronal vesicles are labeled with specific fluorescent proteins to visualize their movement. First, a plasmid coding for a fluorescent protein is incubated with a transfection agent and then introduced into a culture of neurons. Subsequently, images of the neurons are acquired and analyzed to track the vesicles' movement.

The investigation of membrane trafficking is crucial for understanding neuronal functions. Here, we introduce a method for quantifying vesicle motility in neurons. This is a convenient method that can be adapted to the quantification of membrane trafficking in the nervous system.
