The DNA Helix

- L'acide désoxyribonucléique, ou ADN,consiste en deux chaînes de polynucléotides. 
Chacune est faite sous-unités de nucléotides,jusqu'à 4 types différents,qui consistent en un groupe phosphate et une base azotée,adénine, A, cytosine, C, guanine, G, thymine, T,attachées à un sucre désoxyribose. 
Cet arrangement crée l'ossature de phosphate de sucrede la structure de l'ADN. 
Sur un brin, à l'extrémité 3',le sucre est orienté avec un groupe hydroxyleexposé au troisième atome de carbone,qui est prêt à réaliser une liaison covalenteà une autre molécule de sucre dans la chaînesur le 5ème atome carbone, par une liaison phosphodiester. 
Sur le brin opposé antiparallèle,chaque base est appariée de manière unique,C avec G et A avec T,grâce aux différence dans le nombre de liaisons hydrogènesqui se forment entre elles. 
Ce brin complémentaire se termine par un groupe phosphatesur la fin 5'. 
Une telle polarité chimique garantit un paquet efficace,enroulant les deux brins l'un autour de l'autrepour former une double hélice 3D avec 10 paires de baseà chaque tour complet.

 L’acide désoxyribonucléique, ou ADN, est le matériel génétique responsable de la transmission des traits de génération en génération dans tous les organismes et la plupart des virus. L’ADN est composé de deux brins de nucléotides qui s’enroulent l’un autour de l’autre pour former une double hélice. La découverte de la structure de l’ADN s’est produite progressivement sur près d’un siècle, représentant l’un des récits les plus célèbres et captivants de l’histoire de la science.
 
 <h4>La structure de l’ADN en détail</h4>
 
 Chaque brin d’ADN se compose de sous-unités appelées nucléotides qui contiennent le sucre désoxyribose, un groupe phosphate et l’une des quatre bases contenant de l’azote: adénine (A), guanine (G), cytosine (C) et thymine (T). L’adénine et la guanine sont membres d’une plus grande classe de produits chimiques qu’on appelle les purines, qui contiennent tous des structures à deux cycles. La cytosine et la thymine appartiennent à un groupe de structures à un seul cycle nommées pyrimidines. 
 
 Les nucléotides adjacents dans le même brin sont liés de façon covalente par des liaisons phosphodiester. Les deux brins de nucléotides sont maintenus ensemble par des liaisons hydrogène, dans lesquelles les adénines dans un brin s’associent à des thymines à la même position dans l’autre brin, et les cytosines dans un brin s’associent avec des guanines à la même position dans l’autre brin. Ces liaisons hydrogène sont rendues possible par l’arrangement antiparallèle des deux brins d’ADN, dans lequel les extrémités 5’ et 3’ des brins sont orientées dans des directions opposées. Sans cet arrangement, les nucléotides seraient dans la mauvaise position pour former des liaisons hydrogène entre les brins.
 
 Les deux brins de la molécule d’ADN sont étroitement enroulés dans une structure en forme de ressort appelée double hélice. Cependant, la double hélice n’est pas parfaitement symétrique. Au lieu de cela, il y a des sillons qui se produisent régulièrement dans la structure. Le sillon principal se produit à l’endroit où les squelettes sucre-phosphate sont relativement éloignés. Cet espace donne l’accès à des protéines liant l’ADN, telles que des facteurs de transcription. Le sillon mineur, en revanche, se produit lorsque les squelettes sucre-phosphate sont proches les uns des autres. Relativement peu de protéines se lient à l’ADN par l’intermédiaire du sillon mineur.
 
 <h4>La découverte de la structure de l’ADN : une brève histoire</h4>
 
 L’histoire de la découverte de la structure de l’ADN commence en 1869 lorsque le scientifique suisse Friedrich Miescher a découvert une substance qu’il a appelée “ nucléine ”. Au cours du processus d’extraction de protéines à partir de globules blancs, Miescher a trouvé une substance inattendue qui avait une teneur en phosphore relativement élevée. Il ne savait pas ce que c’était, mais il soupçonnait que cela pourrait être important du point de vue biologique. Miescher avait raison, mais il a fallu des décennies à la communauté scientifique pour apprécier pleinement ses idées.
 
 La prochaine découverte cruciale a été réalisée par le biochimiste russe Phoebus Levene. En 1919, Levene proposa que la nucléine, alors connue sous le nom d’acide nucléique, soit composée de chaînes de molécules qu’il appela polynucléotides. La proposition de Levene découlait de ses recherches sur la levure, dans lesquelles il a constaté que les nucléotides individuels étaient composés d’un groupe phosphate, d’un sucre et d’une base contenant de l’azote. Bien que le modèle du polynucléotide de Levene ait été correct à bien des égards, on ne savait toujours pas comment les bases étaient disposées dans la molécule d’ADN.
 
 Le biochimiste autrichien Erwin Chargaff a élargi les travaux de Levene. En travaillant à la fin des années 1940, Chargaff a fait une constatation clé : la quantité d’adénine dans l’ADN est toujours à peu près égale à la quantité de thymine, et la quantité de guanine est toujours à peu près égale à la quantité de cytosine. Ce modèle est devenu connu sous le nom de la règle de Chargaff et c’était un élément de preuve essentiel qui a permis l’élucidation finale de la structure de l’ADN.
 
 Au début des années 1950, le biologiste américain James Watson et le physicien anglais Francis Crick faisaient la course à leur principal rival, l’américain Linus Pauling, pour découvrir la structure tridimensionnelle de l’ADN. S’appuyant sur le travail de Chargaff, ils ont utilisé la connaissance de la physique, des mathématiques et de la chimie pour construire des modèles physiques de l’ADN. Mais ils n’ont pas réussi jusqu’à ce qu’ils reçoivent un morceau crucial de données: une “ photographie ” aux rayons X de l’ADN qui indiquait en détail sa structure double-hélicoïdale. Cette photographie était les données inédites de la physicienne Rosalind Franklin et elle a été donnée à Watson et Crick à l’insu de Franklin. Watson et Crick ont publié leur description de la structure de l’ADN en 1953 et, avec Maurice Wilkins (un collègue de Franklin), ils ont remporté le prix Nobel de physiologie ou de médecine en 1962 pour cette découverte. Malheureusement, Franklin est morte en 1958 et elle était donc inéligible pour un prix Nobel.
