Membrane Lipids

Trois types de lipides sont présents dans la membrane : les phosphoglycérides, les sphingolipides et les stérols.
Les phosphoglycérides, également connus sous le nom de glycérophospholipides, contiennent un squelette glycérol avec un groupe phosphate hydrophile et deux chaînes acyles grasses hydrophobes, généralement de 16 à 22 atomes de carbone.
Certains des phosphoglycérides les plus courants présents dans la membrane cellulaire sont la phosphatidylcholine, la phosphatidyléthanolamine, la phosphatidylsérine et le phosphatidylinositol - formés par la fixation de la choline, de l’éthanolamine, de la sérine et de l’inositol au groupe phosphate.
Les sphingolipides sont des lipides qui contiennent un squelette de sphingosine. La sphingosine est un alcool aminé doté d’une longue chaîne d’hydrocarbures insaturés. 
Le sphingolipide le plus simple est le céramide, formé lorsqu’une molécule d’acide gras se fixe au groupe amino de la sphingosine.
Le groupe hydroxyle restant des céramides peut former des esters avec d’autres groupes chimiques tels que la phosphocholine ou les glucides, formant respectivement de la sphingomyéline ou des glycolipides.
Le troisième type de lipides membranaires, les stérols, tels que le cholestérol, ne peuvent pas former indépendamment une bicouche lipidique. Au lieu de cela, ils s’intercalent au sein des bicouches de phospholipides et agissent comme un tampon de fluidité pour la membrane.

Les lipides sont un composant essentiel de toutes les membranes biologiques. La teneur moyenne en lipides des membranes des mammifères est de 50 %, bien qu'elle puisse descendre jusqu'à 20 % dans la membrane mitochondriale interne ou atteindre jusqu'à 80 % dans la gaine de myéline présente autour des cellules nerveuses.

La phosphatidylcholine, la phosphatidyléthanolamine, la phosphatidylsérine et la sphingomyéline sont les phospholipides les plus présentes dans les membranes des mammifères. Au pH physiologique, la phosphatidylsérine est chargée négativement, tandis que les trois autres restent neutres. Dans les lipides membranaires, le nombre d'atomes de carbone présents dans les acides gras et leur degré de saturation peuvent varier. Les acides gras peuvent être entièrement saturés sans doubles liaisons, monoinsaturés avec une seule double liaison ou polyinsaturés avec plusieurs doubles liaisons.

Les glycolipides constituent une classe physiologiquement importante de lipides membranaires qui sont toujours présents du côté externe ou extracellulaire de la membrane. Chez les eucaryotes, on les retrouve à la fois dans la membrane plasmique et dans les membranes intracellulaires. Le galactocérébroside, un glycolipide, existe en abondance dans les gaines de myéline avec la sphingomyéline. Les gangliosides, les glycolipides les plus complexes, sont chargés négativement en raison de la présence d'acide sialique et peuvent constituer jusqu'à 10 % du total des lipides dans les membranes des cellules nerveuses. Les gangliosides sont étudiés pour leur rôle dans les maladies virales et génétiques. Le ganglioside GM1 agit comme un récepteur de surface cellulaire pour la toxine du choléra, tandis qu'une accumulation excessive de gangliosides GM2 est observée dans la maladie de Tay-Sachs, une maladie génétique.

Une autre classe de lipides membranaires est celle des stérols. Les stérols spécifiques aux plantes constituent un composant important de la membrane plasmique végétale et peuvent représenter 30 à 50 % du total des lipides membranaires. Dans les membranes eucaryotes, les cholestérols sont également présents, constituant près de 50 % des lipides totaux. La structure du cholestérol dans les membranes cellulaires humaines est légèrement différente de celle de l'ergostérol, un constituant lipidique commun des membranes fongiques.

Membrane lipids give biological membranes their structure and help set their properties. In mammalian cells, lipids make up about 50% of the membrane on average. The amount can be much lower in the inner mitochondrial membrane or much higher in the myelin sheath around nerve cells.

Several phospholipids are common in mammalian membranes. These include phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, and sphingomyelin. At physiological pH, phosphatidylserine carries a negative charge, while the other three are neutral.

The fatty acids in membrane lipids can differ in carbon length and in saturation. Some are fully saturated, which means they have no double bonds. Others are monounsaturated with one double bond, or polyunsaturated with multiple double bonds.

Glycolipids are an important membrane lipid class that stays on the outer, or extracellular, side of the membrane. In eukaryotes, they are found in both the plasma membrane and intracellular membranes. Galactocerebroside is a simple glycolipid found in large amounts in myelin sheaths along with sphingomyelin.

Gangliosides are the most complex glycolipids. They are negatively charged because they contain sialic acid, and they can make up as much as 10% of the total lipids in nerve cell membranes. Their roles are also studied in viral and genetic diseases. GM1 acts as a cell surface receptor for cholera toxin, and excess GM2 ganglioside is seen in Tay-Sachs disease.

Sterols are another major class of membrane lipids. Plant-specific sterols can make up 30% to 50% of the total lipids in the plant plasma membrane. In eukaryotic membranes, cholesterol is also common and can account for almost 50% of the total lipids. Cholesterol in human cell membranes is slightly different from ergosterol, which is a common lipid in fungal membranes.
