Mechanisms of Membrane-bending

Une membrane a une courbure positive lorsqu’elle se plie vers l’intérieur ou le côté cytoplasmique. En revanche, une membrane a une courbure négative si elle se plie vers le côté externe ou extracellulaire.
Il existe de multiples mécanismes qui peuvent provoquer la flexion de la membrane.
La flexion membranaire induite par les lipides est due à l’agrégation asymétrique de lipides spécifiques dans la membrane.
Les lipides avec de grands groupes de tête induisent une courbure membranaire positive, tandis que les lipides avec de petits groupes de tête produisent une courbure membranaire négative.
L’échafaudage protéique est un autre mécanisme par lequel les protéines déforment la membrane ou stabilisent une membrane déjà déformée.
Dans un mécanisme différent, les domaines protéiques hydrophobes sont insérés de manière réversible dans une couche de la membrane pour augmenter sa surface par rapport à l’autre couche. Cette asymétrie force la membrane à se plier.
Enfin, les forces de poussée et de traction générées par l’assemblage et le désassemblage du cytosquelette et de ses protéines motrices associées peuvent également plier la membrane pour former des projections, comme les filopodes dans les cellules épithéliales.

Les membranes vivantes sont flexibles en raison de leur nature fluide en mosaïque ; cependant, leur courbure sous différentes formes est un processus actif régulé par les lipides et les protéines spécifiques. La courbure de la membrane peut être transitoire comme dans les vésicules ou stable pendant une longue période comme dans les microvillosités. Les cellules régulent la taille, l'emplacement et la durée de la courbure de la membrane.

La courbure de la membrane peut se produire en raison de changements intrinsèques dans la composition lipidique ou d'une association extrinsèque avec différentes protéines. Les protéines impliquées dans la création d’une asymétrie membranaire ou dans le regroupement de lipides spécifiques jouent un rôle essentiel dans la courbure membranaire induite par les lipides. La forme des protéines intégrales peut également affecter la courbure de la membrane. Par exemple, les protéines transmembranaires de forme conique ou conique-inversée produisent une courbure de la membrane. L'insertion de protéines amphipathiques, comme l'epsine, dans l'une des couches entraîne également la courbure de la membrane.

Les protéines courbes comme les protéines dimères Bin/amphiphysine/Rvs ou BAR créent une courbure membranaire positive, tandis que la protéine BAR inverse ou I-BAR induit une courbure négative dans la membrane. Les protéines BAR insèrent également leur domaine hydrophobe dans la membrane, ce qui augmente davantage la courbure de la membrane. Outre les mécanismes bien établis, l’encombrement des protéines est un mécanisme récemment découvert dans lequel la pression latérale générée par les interactions protéine-protéine entraîne la courbure de la membrane. Les cytosquelettes jouent également un rôle important dans la courbure de la membrane et les forces qu'ils génèrent sont responsables de la morphologie de certains organites comme le réticulum endoplasmique et l'appareil de Golgi.

Les organites cellulaires comme le réticulum endoplasmique, l'appareil de Golgi et les mitochondries ont des membranes complexement repliées ; ces pliages sont indispensables à leur fonctionnement. La courbure de la membrane est également nécessaire à la scission membranaire, à la fusion membranaire et à l'activation de l'enzyme de détection de courbure membranaire. Les mutations dans les gènes qui produisent des protéines de courbure -membranaire  telles que la myotubularine, l'amphiphysine-2 et la dynamine-2 peuvent conduire à un groupe de troubles neuromusculaires appelés myopathies centronucléaires.

Cell membranes can bend into many shapes because they are flexible and made of a fluid mosaic. That bending can be temporary, as in vesicles, or long-lasting, as in microvilli. Cells control the size, place, and time of membrane curvature.

Membrane bending can start inside the membrane itself or from proteins attaching to it. Changes in lipid makeup can create curvature, and proteins can help by making the membrane asymmetric or by clustering certain lipids. The shape of integral proteins also matters. Conical or inverse-conical transmembrane proteins can push the membrane to bend.

Some proteins bend membranes by inserting part of themselves into one layer of the membrane. Amphipathic proteins, such as epsin, do this and promote curvature. BAR proteins, short for Bin/amphiphysin/Rvs proteins, create positive curvature, while I-BAR proteins create negative curvature. BAR proteins can also add to bending by placing a hydrophobic domain into the membrane.

Protein crowding is another way membranes bend. In this case, proteins packed close together create lateral pressure, and that pressure drives curvature. The cytoskeleton also helps shape membranes. Its forces are important for the form of organelles such as the endoplasmic reticulum and Golgi apparatus.

The folded membranes of the endoplasmic reticulum, Golgi apparatus, and mitochondria are important for their function. Membrane bending also supports membrane scission, membrane fusion, and the activation of enzymes that sense curvature. Changes in genes for membrane-bending proteins, including myotubularin, amphiphysin-2, and dynamin-2, can cause centronuclear myopathies, a group of neuromuscular disorders.
