Dans la synthèse d’esters maloniques, les β-diesters portant des protons α acides en présence d’une base et d’un halogénure d’alkyle génèrent des acides acétiques substitués via des intermédiaires d’esters maloniques alkylés.
Initialement, la base élimine le proton α du diester pour générer un ion énolate nucléophile qui est fortement stabilisé par trois structures de résonance. 
L’alkylation du carbone anionique par un mécanisme SN2 produit un intermédiaire d’ester alkylmalonique. Celui-ci subit une hydrolyse suivie d’une acidification pour donner le β-diacide. 
Le β-diacide, à haute température, subit un processus concerté qui expulse le CO2 pour produire un énol. Celui-ci se tautomérise rapidement jusqu’à la forme céto stable de l’acide acétique substitué. 
Comme la réaction comprend l’élimination d’un seul proton, l’acide résultant est monosubstitué. 
Si, toutefois, l’intermédiaire monoalkylé est soumis au second processus d’alkylation, il en résultera un acide disubstitué.

Malonic ester synthesis is a way to make alpha-substituted carboxylic acids from beta-diesters such as diethyl malonate and alkyl halides. It builds a new carbon-carbon bond and then turns the product into a carboxylic acid.

The reaction starts by removing the acidic alpha hydrogen from the beta-diester. This forms a doubly stabilized enolate ion, which is a nucleophile, meaning it can attack an electron-poor partner.

The enolate reacts with the alkyl halide by an SN2 mechanism. That step gives an alkylated malonic ester intermediate with the new carbon-carbon bond in place.

Next, aqueous acid or base hydrolyzes both ester groups. This forms a 1,3-dicarboxylic acid. When heated, that diacid is unstable and loses CO2 through a cyclic six-membered transition state.

The decarboxylation produces an enol, and the enol then tautomerizes to the more stable keto form. The final product is a monosubstituted carboxylic acid. If the deprotonation and alkylation steps are repeated before hydrolysis and decarboxylation, a disubstituted carboxylic acid can be made instead.
