Les cadhérines sont une superfamille de molécules d’adhésion cellulaire dépendantes du calcium, ou CAM.
Toutes les cadhérines ont une structure de base comprenant un petit domaine cytosolique C-terminal, un seul domaine transmembranaire et plusieurs domaines cadhérines extracellulaires ou EC.
Ces domaines EC sont reliés les uns aux autres par des régions charnières flexibles, chacune avec trois poches de liaison au calcium.
Lorsque les ions calcium se lient, la cadhérine devient rigide, ce qui permet la liaison cis et trans à d’autres cadhérines.
Dans la liaison cis, les cadhérines d’une même cellule peuvent se lier les unes aux autres pour former des grappes, tandis que l’interaction des domaines EC des cadhérines sur les cellules adjacentes entraîne une transliaison.
Le domaine EC N-terminal, le plus éloigné de la membrane, appelé EC1, a une structure en bouton et en poche. Lors de la transliaison, le bouton d’un EC1 s’insère dans la poche d’un autre EC1 sur la cellule adjacente.
Cela fait que les cadhérines d’un type lient préférentiellement les cadhérines du même type, une liaison homophile.
Bien que l’interaction entre une seule paire de cadhérines soit relativement faible, les grappes de cadhérines peuvent former une trans-liaison collectivement forte. De tels complexes forment des jonctions cellulaires d’ancrage.

Structure of Cadherins

Les cadhérines ont été l'une des premières molécules d'adhésion cellulaire découvertes ; le terme « cadhérines » est basé sur leurs propriétés adhérentes dépendantes du calcium. Les premières cadhérines découvertes sur les cellules épithéliales, neuronales et placentaires étaient respectivement nommées E-cadhérine, P-cadhérine et N-cadhérine. Ces cadhérines classiques partagent des similitudes de séquence et de structure. D'autres cadhérines, notamment celles impliquées dans la signalisation cellulaire, sont regroupées en cadhérines non classiques. Cette diversité de cadhérines provient de plusieurs gènes de cadhérines dans le génome et d'un épissage alternatif d'ARN au cours de la transcription.

Cadhérines dans les Jonctions Cellulaires

Les cadhérines sont des protéines transmembranaires possédant de multiples domaines de liaison extracellulaires. Leur domaine cytosolique lie les protéines de liaison qui se connectent au cytosquelette. Les cadhérines se regroupent sur la membrane plasmique par liaison cis, formant de grands complexes multiprotéiques appelés jonctions cellulaires. Alors que les cadhérines classiques telles que la E-cadhérine forment des jonctions adhérentes, les cadhérines non classiques comme la desmogléine et la desmocolline forment des jonctions desmosomales. Ces jonctions cellulaires ont pour fonction de maintenir les cellules ensemble et de fournir une résistance mécanique aux tissus, tels que l'épithélium.

Importance Clinique des Cadhérines

Les cadhérines jouent un rôle essentiel dans l'adhésion cellulaire, la formation des tissus et la communication intercellulaire. Par conséquent, tout changement dans leur structure, leur expression et leur stabilité peut se manifester par des conditions cliniques. Lorsque les cellules perdent le contact avec les cellules voisines, elles peuvent se libérer du tissu et envahir d’autres tissus. La perte d'adhésion cellulaire due à des mutations dans les gènes de la cadhérine est associée à la malignité des tumeurs, telles que celles observées dans les cellules malignes du sein. Les agents pathogènes utilisent également les cadhérines comme récepteurs pour s'attacher et envahir la cellule hôte. Par exemple, la cadhérine CDHR3 est fortement exprimée dans l’épithélium pulmonaire et est utilisée comme récepteur par le rhinovirus de classe C (RV-C) responsable du rhume.

Cadherins are cell adhesion proteins that help cells stick together and build tissues. Their name comes from their calcium-dependent binding. Early examples were named E-cadherin, P-cadherin, and N-cadherin after the epithelial, placental, and neuronal cells where they were first found.

Classical cadherins share similar sequences and structures. Other cadherins, including those involved in cell signaling, are grouped as non-classical cadherins. This diversity comes from multiple cadherin genes in the genome and from alternative RNA splicing during transcription.

Cadherins are transmembrane proteins. They have several extracellular binding domains and a cytosolic domain inside the cell. The cytosolic region binds linker proteins, which connect cadherins to the cytoskeleton. Cadherins also cluster on the plasma membrane through cis-binding, which helps form large protein complexes called cell junctions.

Classical cadherins such as E-cadherin form adherens junctions. Non-classical cadherins such as desmoglein and desmocollin form desmosomal junctions. These junctions hold cells together and give mechanical strength to tissues such as epithelium.

Cadherins are important for cell adhesion, tissue formation, and communication between cells. Changes in their structure, expression, or stability can lead to disease. When cells lose contact with neighboring cells, they may break free from the tissue and invade other tissues. Mutations in cadherin genes are linked to tumor malignancy, including in breast cancer cells. Some pathogens also use cadherins as receptors to attach to and enter host cells. One example is CDHR3, which is highly expressed in lung epithelium and is used by class-C rhinovirus, or RV-C, the virus that causes the common cold.
