Anaphase Promoting Complex

La réussite de l’anaphase dépend également de l’activité d’une ubiquitine ligase appelée complexe favorisant l’anaphase ou cyclosome APC/C.
Au cours de la transition vers l’anaphase, la protéine Cdc20 se lie à l’APC/C et l’active. L’APC/C activée ubiquitine les protéines régulatrices, les S-cyclines et les M-cyclines, les ciblant ainsi pour la dégradation protéasomale.
La dégradation des cyclines inhibe l’activité de la plupart des Cdk. L’absence d’activité kinase dépendante des cyclines permet aux phosphatases de déphosphoryler la plupart des substrats Cdk à l’intérieur de la cellule, ce qui est nécessaire à la réalisation de l’anaphase.

La destruction progressive de protéines spécifiques est nécessaire à la progression et à l’achèvement du cycle cellulaire. Ces protéines sont ubiquitinées par les ubiquitine ligases puis détruites par le protéasome. Le SCF (Skp1/Cullin/F-box) et le complexe favorisant l'anaphase (APC) sont deux ubiquitine ligases importantes impliquées dans la progression du cycle cellulaire. Alors que le SCF est actif tout au long du cycle cellulaire, l’APC est activé pendant la transition métaphase-anaphase. Cdc20 ou Cdh1 se lie à l'APC et l'active, entraînant respectivement la formation de complexes APC^Cdc20 et APC^Cdh1. Cdc20 et Cdh1 aident également APC à identifier ses substrats.

APC^Cdc20 joue un rôle essentiel dans la transition métaphase-anaphase. Les chromatides sœurs sont maintenues ensemble par des cohésines. Les anneaux de cohésine doivent être brisés par l'enzyme séparase afin que les chromatides sœurs puissent se déplacer vers les pôles opposés pendant l'anaphase. Cependant, l’activité séparase est inhibée par la protéine sécurine. APC^Cdc20 ubiquitine la sécurine, conduisant à sa dégradation protéasomale. Cela conduit à l’activation ultérieure de la séparase. APC^Cdc20 ubiquitine également les cyclines mitotiques, ce qui à son tour inactive les kinases dépendantes des cyclines ou Cdks. L'inactivation des Cdk et la déphosphorylation ultérieure des substrats Cdk sont nécessaires à l'achèvement de la division cellulaire.
