The Central Dogma

Le dogme central de la biologie moléculaire stipule que l’information codée dans l’ADN est transférée à l’ARN, qui dirige ensuite la synthèse des protéines en fonction de ces instructions.
Tout d’abord, lors de la transcription, l’ADN est utilisé comme matrice pour synthétiser l’ARN messager ou ARNm.
L’ARNm représente une copie du brin codant de l’ADN, sauf que les thymines sont remplacées par des uraciles.
Ensuite, l’ARNm est traduit en une chaîne d’acides aminés par un ribosome.
Ici, des groupes de trois nucléotides sur l’ARNm, également appelés codons, se lient à des séquences complémentaires ou à des anticodons sur les molécules d’ARN ou d’ARNt de transfert.
Chaque ARNt est attaché à un acide aminé particulier, en fonction du codon spécifique.
Par exemple, le codon CCA se lie à un ARNt attaché à la proline, tandis que l’AGC se lie à un ARNt attaché à la sérine.
De cette façon, le code génétique spécifie l’ordre des acides aminés dans le polypeptide résultant, qui est ensuite traité pour devenir une protéine fonctionnelle.

L'ARN est le chaînon manquant entre l'ADN et les protéines

Au début des années 1900, les scientifiques ont découvert que l’ADN stockait toutes les informations nécessaires aux fonctions cellulaires et que les protéines remplissaient la plupart de ces fonctions. Cependant, les mécanismes de conversion de l’information génétique en protéines fonctionnelles sont restés inconnus pendant de nombreuses années. Initialement, on pensait qu’un seul gène était directement converti en sa protéine codée. Deux découvertes cruciales sur les cellules eucaryotes ont remis en question cette théorie. Premièrement, la production de protéines n’a pas lieu dans le noyau. Deuxièmement, l’ADN n’est pas présent en dehors du noyau. Ces découvertes ont déclenché la recherche d’une molécule intermédiaire reliant l’ADN à la production de protéines. Cette molécule intermédiaire s’est avérée être de l’ARN.

L'ARN est synthétisé dans le noyau en utilisant l'ADN comme matrice. L'ARN nouvellement synthétisé a une séquence similaire à celle du brin d'ADN, sauf que la thymine est remplacée par l'uracile. Chez les eucaryotes, ce transcrit primaire est ensuite traité, en supprimant les régions non codantes des protéines, en coiffant l'extrémité 5' et en ajoutant une queue poly-A 3' pour créer un ARNm mature, qui est ensuite exporté vers le cytoplasme.

Le Code Génétique est Redondant

Les protéines sont créées à partir de 20 acides aminés chez les eucaryotes. La combinaison de quatre nucléotides en ensembles de trois fournit 64 (43) codons possibles. Cela signifie que plus d'un codon peut coder pour un acide aminé individuel. Ainsi, le code génétique est dit redondant ou dégénéré. Souvent, mais pas toujours, les codons qui spécifient les mêmes acides aminés ne diffèrent que par le troisième nucléotide du triplet. Par exemple, les codons GUU, GUC, GUA et GUG représentent tous l'acide aminé valine. Cependant, AUG est le seul codon qui représente l’acide aminé méthionine. Le codon AUG est également le codon où commence la synthèse des protéines et est donc appelé codon d'initiation. La redondance du système minimise les effets néfastes des mutations. Par exemple, une modification de la troisième position du codon peut ne pas nécessairement modifier l'acide aminé codé, n'entraînant ainsi aucun changement dans la fonctionnalité de la protéine.

Le Code Génétique est Universel

À quelques exceptions près, la plupart des organismes procaryotes et eucaryotes utilisent le même code génétique pour la synthèse des protéines. Cette universalité du code génétique a permis des avancées dans la recherche scientifique dans le domaine de l’agriculture et de la médecine. Par exemple, l’insuline humaine peut désormais être fabriquée à grande échelle par des bactéries grâce à la technologie de l’ADN recombinant. La technologie de l’ADN recombinant implique l’utilisation de matériel génétique provenant de différentes espèces. Les gènes codant pour l'insuline humaine sont associés à l'ADN bactérien et insérés dans une cellule bactérienne. La cellule bactérienne effectue ensuite la transcription et la traduction pour produire la protéine insuline humaine, qui peut traiter le diabète chez les patients.
