Replication in Eukaryotes

La réplication de l’ADN dans les chromosomes eucaryotes commence à plusieurs origines de réplication, qui sont identifiées et liées par le complexe de reconnaissance d’origine, ou ORC.
L’ORC recrute ensuite des hélicases pour dérouler l’ADN, produisant une bulle de réplication avec deux fourches de réplication.
Les deux fourches se déplacent dans des directions opposées et perturbent les nucléosomes devant elles. Ces nucléosomes sont ensuite réassemblés sur les brins filles, en maintenant la structure de la chromatine.
À chaque fourche, les amorces d’ARN fournissent le site où l’ADN polymérase permet d’allonger le brin principal et les fragments d’Okazaki du brin retardé.
Une enzyme RNase élimine ensuite ces amorces, et l’ADN polymérase comble les lacunes. Enfin, l’ADN ligase scelle les fragments ensemble.
Cependant, lorsque la dernière amorce est retirée du brin retardé à l’extrémité du chromosome linéaire, elle produit un étirement en surplomb de l’ADN matrice.
Une enzyme appelée télomérase prolonge cette élongation avec de l’ADN non codant pour empêcher la perte d’ADN codant lors des cycles de réplication ultérieurs.
La réplication se poursuit jusqu’à ce que les bulles de réplication adjacentes fusionnent et que le chromosome entier soit dupliqué.

Dans les cellules eucaryotes, la réplication de l’ADN est hautement conservée et étroitement régulée. Plusieurs chromosomes linéaires doivent être dupliqués avec une haute fidélité avant la division cellulaire. De nombreuses protéines remplissent donc des rôles spécialisés dans le processus de réplication. La réplication se déroule en trois phases : initiation, élongation et terminaison, et se termine par deux ensembles complets de chromosomes dans le noyau.

De Nombreuses Protéines Orchestrent la Réplication à l’Origine

La réplication eucaryote suit bon nombre des mêmes principes que la réplication de l'ADN procaryote, mais comme le génome est beaucoup plus grand et que les chromosomes sont linéaires plutôt que circulaires, le processus nécessite plus de protéines et présente quelques différences clés. Premièrement, contrairement aux procaryotes, la réplication chez les eucaryotes se produit simultanément à plusieurs origines de réplication le long de chaque chromosome. Les protéines initiatrices reconnaissent et se lient à ces origines et recrutent des protéines hélicase pour dérouler la double hélice de l'ADN. A chaque point d'origine, deux fourches de réplication se forment. Primase ajoute ensuite de courtes amorces d'ARN aux simples brins d'ADN, qui servent de point de départ à l'ADN polymérase pour se lier et commencer à copier la séquence. L'ADN ne peut être synthétisé que dans la direction 5' vers 3', de sorte que la réplication des deux brins à partir d'une seule fourche de réplication se déroule dans deux directions différentes. Le brin principal est synthétisé en continu, tandis que le brin retardateur est synthétisé en courtes séquences de 100 à 200 paires de bases de longueur, appelées fragments d'Okazaki. Une fois la majeure partie de la réplication terminée, les enzymes RNase éliminent les amorces d’ARN, l’ADN polymérase comble les lacunes et l’ADN ligase comble les lacunes du nouveau brin.

Répartir le Travail de Réplication entre les Polymérases

La charge de travail de copie de l’ADN chez les eucaryotes est répartie entre plusieurs types différents d’enzymes ADN polymérase. Les grandes familles d’ADN polymérases dans tous les organismes sont classées en fonction de la similitude de leurs structures protéiques et de leurs séquences d’acides aminés. Les premières familles découvertes ont été appelées A, B, C et X, les familles Y et D étant identifiées plus tard. Les polymérases de la famille B chez les eucaryotes comprennent Pol α, qui fonctionne également comme une primase au niveau de la fourche de réplication, et Pol δ et ε, les enzymes qui effectuent l'essentiel du travail de réplication de l'ADN sur les brins principaux et retardés de la matrice, respectivement. D'autres ADN polymérases sont responsables de tâches telles que la réparation des dommages à l'ADN, la copie de l'ADN mitochondrial et plastidique et le comblement des lacunes dans la séquence d'ADN sur le brin en retard après le retrait des amorces d'ARN.

Les Télomères Protègent les Extrémités des Chromosomes de la Dégradation

Les chromosomes eucaryotes étant linéaires, ils sont susceptibles de se dégrader aux extrémités. Pour protéger les informations génétiques importantes contre tout dommage, les extrémités des chromosomes contiennent de nombreuses répétitions non codantes d’ADN riche en G hautement conservé, les télomères. Un court surplomb 3' simple brin à chaque extrémité du chromosome interagit avec des protéines spécialisées qui stabilisent le chromosome dans le noyau. En raison de la manière dont le brin retardateur est synthétisé, une petite quantité de l’ADN télomérique ne peut pas être répliquée à chaque division cellulaire. En conséquence, les télomères raccourcissent progressivement au cours de nombreux cycles cellulaires et peuvent ainsi être mesurés comme un marqueur du vieillissement cellulaire. Certaines populations de cellules, telles que les cellules germinales et les cellules souches, expriment la télomérase, une enzyme qui allonge les télomères, permettant à la cellule de subir davantage de cycles cellulaires avant que les télomères ne raccourcissent.
