Cellular Differentiation

Au cours de la différenciation cellulaire, les cellules non spécialisées acquièrent des caractéristiques, telles que la forme, la taille ou la composition des protéines membranaires, pour remplir des fonctions distinctes dans le corps.
Un ovule fécondé ou zygote est très prolifératif et totipotent, c’est-à-dire qu’il peut se différencier pour former tous les types de cellules.
Au fur et à mesure que le zygote subit une série de divisions, les cellules filles perdent leur totipotence et se différencient pour former des cellules spécialisées dans l’embryon, telles que les cellules musculaires, les neurones, l’épiderme de la peau, les cellules cardiaques et les cellules hépatiques.
Bien que toutes ces cellules du corps contiennent le même génome, elles expriment certains gènes tout en désactivant d’autres. Cela leur permet de synthétiser les protéines nécessaires à l’exécution de fonctions spécifiques aux tissus.

Tout commence par un seul ovule fécondé qui donne naissance à une vaste gamme de types cellulaires, tels que les cellules nerveuses, musculaires et épithéliales qui caractérisent l'adulte. Tout au long du développement et de l'âge adulte, la différenciation cellulaire amène les cellules à assumer leur morphologie et leur physiologie finales. La différenciation est le processus par lequel les cellules non spécialisées deviennent spécialisées pour accomplir des fonctions distinctes. 

 Un zygote est une cellule souche totipotente avec un potentiel élevé de différenciation qui peut former tous les types cellulaires et donner naissance à un nouvel organisme pluricellulaire complet. Au fur et à mesure que le zygote subit des divisions cellulaires successives, il génère des cellules souches pluripotentes (CSP) avec un potentiel de différenciation plus restreint. Les CSP donnent naissance à quelques cellules souches adultes qui se divisent davantage et se différencient de manière terminale. Lorsqu'une cellule se différencie, elle peut subir d'importants changements de taille, de forme, d'activité métabolique et de fonction globale.  

 À partir de l'œuf fertilisé, toutes les cellules du corps contiennent le même ADN, mais chaque type de cellule ne "lit" que les parties de l'ADN qui sont pertinentes pour sa fonction. Plusieurs facteurs tels que les hormones ou les molécules de signalisation déclenchent la différenciation cellulaire et, en réponse, activent l'expression de gènes spécifiques tout en en désactivant d'autres. Le principal mécanisme par lequel les gènes sont activés ou désactivés est à travers les facteurs de transcription. Un facteur de transcription est une classe de protéines qui se lient à des gènes spécifiques sur la molécule d'ADN et qui favorisent ou inhibent leur transcription. L'expression de gènes spécifiques médie la synthèse des protéines nécessaires à une fonction cellulaire particulière, aidant ainsi à la différenciation cellulaire.

Cellular differentiation turns unspecialized cells into specialized cells with specific jobs. This process helps a complex organism, such as a human, develop from a single fertilized egg into a body with many cell types. Nerve cells, muscle cells, and epithelial cells are examples of the final cell types that appear during development.

A zygote is the first cell after fertilization. It is a totipotent stem cell, which means it has very high differentiation potential and can form all cell types and even produce a whole new multicellular organism. As the zygote divides, it gives rise to pluripotent stem cells, or PSCs, which have a more limited ability to differentiate. PSCs then produce a few adult stem cells that keep dividing until they become more terminally differentiated.

As cells differentiate, they can change in size, shape, metabolic activity, and overall function. Even though the body’s cells contain the same DNA, each cell type only reads the parts of the DNA that are relevant to its role. This selective reading helps each cell become suited to its own job.

Hormones and signaling molecules can trigger differentiation. In response, specific genes are turned on while others are turned off. Transcription factors, which are proteins that bind to specific genes on DNA, control this switch by promoting or inhibiting transcription. The expression of the right genes leads to the proteins needed for a particular cell function and supports cellular differentiation.
