Une méthodologie analytique comporte quatre niveaux en cascade : technique, méthode, procédure et protocole.
Une technique est un principe scientifique qui rationalise un phénomène spécifique à travers des mesures chimiques.
Une méthode est l’adaptation d’une technique d’analyse d’un échantillon particulier d’intérêt.
Une procédure est un ensemble de directives pour effectuer l’analyse.
Un protocole est les directives strictes à respecter lors du détail de la procédure.
Les méthodes analytiques peuvent être classées en deux catégories : classique et instrumentale.
Dans les méthodes classiques, le composant d’intérêt – l’analyte – est séparé du reste de la matrice de l’échantillon par des processus chimiques humides tels que la précipitation, l’extraction ou la distillation.
Alternativement, les méthodes instrumentales sondent les propriétés physiques d’un analyte, telles que la conductivité, le potentiel de l’électrode, l’absorption ou l’émission de lumière et le rapport masse/charge.
Le choix de la méthode analytique est déterminé par les exigences de disponibilité de l’équipement, d’exactitude, de précision, de sensibilité, de sélectivité, de robustesse, de coût, de temps d’analyse et d’échelle de fonctionnement.

Une méthode analytique est choisie en fonction de l'exactitude, de la précision, de la sensibilité, de la sélectivité, de la robustesse, de la durabilité, de l'échelle d'opération, du temps d'analyse, de la disponibilité des équipements et du coût.

Analytical methods follow a clear path from technique to protocol. In analytical chemistry, a technique is the scientific principle used to explain a chemical measurement. A method applies that technique to a sample of interest.

The next steps are procedure and protocol. A procedure gives the directions for carrying out the analysis with an analytical method. A protocol is the detailed set of guidelines for that procedure, and it should be followed closely to get the desired results.

Analytical methods are also grouped into classical and instrumental methods. Classical methods use wet chemical processes, such as precipitation, extraction, or distillation, to separate the analyte, or component of interest, from the matrix, or the rest of the sample. Instrumental methods use instruments to measure physical properties such as conductivity, electrode potential, mass-to-charge ratio, and absorption or emission of light.

The best method depends on several practical factors. These include accuracy, precision, sensitivity, selectivity, robustness, ruggedness, scale of operation, analysis time, equipment availability, and cost. Choosing the right analytical method means matching the test to the goal of the analysis.
