La préparation des échantillons est une étape critique pour obtenir des résultats d’analyse précis. Il comprend trois objectifs principaux : l’extraction, la purification et la concentration.
Tout d’abord, l’extraction consiste à isoler un mélange avec l’analyte d’intérêt à partir d’une matrice d’échantillon. Deuxièmement, si des substances interférentes sont présentes, elles sont éliminées pour obtenir un échantillon pur. Troisièmement, si l’analyte extrait est de faible concentration, il est concentré à un niveau qui peut être détecté avec précision.
Les formes courantes de l’échantillon sont solides, liquides et gazeuses. Les morceaux solides trop gros pour être analysés directement sont pulvérisés par des méthodes physiques telles que le broyage, le broyage et le broyage.

Les échantillons solides en vrac ou de grande taille sont généralement réduits en taille à l'aide de techniques de broyage, d'écrasement ou de mouture pour augmenter la surface de l'échantillon et fournir une analyse plus efficace. Après broyage, l'échantillon est tamisé et mélangé pour garantir une composition uniforme. L'échantillon en poudre est ensuite dissous dans un solvant approprié pour extraire l'analyte de la matrice solide. Le choix du solvant dépend de la nature de l'analyte et des caractéristiques de solubilité des composants de l'échantillon.

Les échantillons liquides qui contiennent des particules solides en suspension ou des précipités sont traditionnellement soumis à une filtration ou à une centrifugation pour séparer les impuretés solides du liquide. La filtration consiste à faire passer le liquide à travers un filtre, tandis que la centrifugation utilise la force centrifuge (rotation) pour séparer les solides de la phase liquide. Si l'analyte est présent en quantités infimes, les échantillons liquides sont soumis à un prétraitement par étapes de concentration pour augmenter la concentration de l'analyte afin d'améliorer la détection. 
La préparation des échantillons est généralement inutile pour l'analyse des analytes gazeux, car l'échantillon de gaz peut être directement introduit dans l'instrument d'analyse.

Sample preparation depends on the state and form of the sample before analysis. It helps make the sample ready for accurate measurement. The main goal is to extract the analyte, which is the substance being measured, and remove parts that could interfere with the analysis.

Solid samples often need the most handling. Large solids are usually ground, crushed, or milled to make them smaller and increase surface area. The sample is then sieved and mixed so it has a uniform composition. After that, the powdered material is dissolved in a suitable solvent to pull the analyte out of the solid matrix. The best solvent depends on the analyte and on how well the sample components dissolve.

Liquid samples may also need treatment before analysis. If they contain suspended solids or precipitates, they are usually filtered or centrifuged to remove the unwanted particles. Filtration works by passing the liquid through a filter. Centrifugation uses spinning force to separate the solid material from the liquid phase.

When the analyte is present in trace amounts, liquid samples may need a concentration step. This increases the analyte level so it is easier to detect. Sample preparation is usually not needed for gaseous analytes because the gas sample can be introduced directly into the analytical instrument.
