Un spectrophotomètre Raman comporte quatre composants clés : une source laser, un système de conservation d’échantillons, un sélecteur de longueur d’onde et un détecteur.
La source laser émet un faisceau focalisé de lumière monochromatique, généralement dans le domaine visible ou proche infrarouge, dont une partie est diffusée par des molécules dans l’échantillon.
Les échantillons peuvent se présenter sous diverses formes, notamment liquides, en solution, solides transparents, en poudre, en granulés ou en gaz.
La lumière diffusée est collectée et dirigée à travers un monochromateur, à l’exclusion de toutes les longueurs d’onde individuelles, sauf certaines longueurs d’onde. 
Les filtres à rejet de bande optique (ou « encoche ») éliminent la lumière du rayonnement laser parasite et de la diffusion de Rayleigh qui peuvent interférer avec le signal Raman.
Les spectromètres Raman à fibre optique, en particulier, utilisent des filtres passe-bande et coupe-bande de haute qualité pour minimiser le rayonnement diffusé par Rayleigh qui atteint le détecteur.
Le détecteur, souvent un dispositif à couplage de charge ou un tube photomultiplicateur, convertit le signal optique en signal électrique, produisant ainsi un spectre Raman.
Les instruments Raman à transformée de Fourier utilisent une source laser à ondes continues ainsi qu’un interféromètre de Michelson au lieu d’un monochromateur, le rayonnement étant focalisé sur une photodiode refroidie pour l’analyse.

Raman Spectroscopy Instrumentation: Overview

Un spectrophotomètre Raman classique comprend une source laser, un système de maintien d'échantillon, un sélecteur de longueur d'onde et un détecteur.

La source laser monochromatique, qui utilise généralement un rayonnement visible ou proche infrarouge, génère un faisceau de lumière hautement focalisé. Cette lumière interagit avec les molécules de l'échantillon, diffusant une partie de la lumière. Les échantillons liquides et gazeux sont généralement testés dans des capillaires en verre ordinaires, tandis que les solides peuvent être analysés sous forme de poudres empaquetées dans des capillaires ou de pastilles de bromure de potassium. La lumière diffusée est collectée à l'aide d'une lentille distincte et focalisée sur l'entrée d'un monochromateur, qui disperse la lumière dans ses fréquences constitutives.

Pour garantir des résultats précis, la sortie est filtrée de manière approfondie pour éliminer le rayonnement laser parasite et la diffusion Rayleigh, qui peuvent interférer avec le signal Raman. Le signal optique est ensuite converti en signal électrique dans le détecteur, souvent un dispositif à couplage de charge (CCD) ou un tube photomultiplicateur, permettant son traitement et sa visualisation sous forme de spectre Raman.

Raman spectroscopy uses a laser, a sample holder, a wavelength selector, and a detector to measure scattered light from a sample. The laser is monochromatic, meaning it has one main wavelength. It usually uses visible or near-infrared light to create a tightly focused beam.

That beam hits the sample and some of the light scatters. Liquid and gas samples are often placed in ordinary glass capillaries. Solid samples can be tested as powders packed into capillaries or as potassium bromide pellets.

The scattered light is collected by a separate lens and sent into a monochromator, which separates the light into its different frequencies. The output is then filtered carefully to remove stray laser light and Rayleigh scattering, which can hide the Raman signal. In fiber-optic Raman instruments, high-quality bandpass and notch filters are often used to reduce Rayleigh-scattered light.

Next, the optical signal is changed into an electrical signal by the detector. A charge-coupled device, or CCD, or a photomultiplier tube may be used. The signal can then be processed and displayed as a Raman spectrum.

Some Raman instruments use a different design called Fourier Transform Raman. In this setup, the monochromator is replaced by a Michelson interferometer, and a continuous-wave laser is used. After filtering, the radiation is focused onto a cooled germanium detector for analysis.
