Differential Staining Technique

La coloration différentielle distingue les micro-organismes en fonction des propriétés de la paroi cellulaire, ce qui facilite l’identification et la classification des bactéries. La coloration de Gram classe les bactéries comme gram-positives ou gram-négatives selon leur capacité à retenir le violet cristallin après la décoloration à l’alcool. Une mince couche de culture microbienne est étalée sur une lame, séchée à l’air et fixée thermiquement. Le frottis est coloré au violet cristallin, lavé à l’eau et traité à l’iode, formant un complexe cristallin violet-iode. L’alcool est utilisé pour décolorer cette tache jusqu’à ce que le ruissellement soit clair, suivi d’un rinçage à l’eau. Les bactéries à Gram positif conservent la tache violette en raison de leur épaisse couche de peptidoglycane. En revanche, les bactéries à Gram négatif perdent la tache violette car l’alcool perturbe la membrane externe et élimine le complexe cristallin violet-iode. La safranine est utilisée pour contre-colorer les bactéries à Gram négatif en rose, tandis que les bactéries à Gram positif restent violettes. La coloration acido-résistante détecte les espèces Mycobacterium , dont les parois cellulaires cireuses et riches en acide mycolique résistent à la décoloration. Les bactéries acido-résistantes conservent le carbol fuchsin, un colorant rouge, tandis que les cellules non acido-résistantes sont généralement contre-colorées au bleu de méthylène.

Technique de coloration de Gram
La coloration de Gram permet de diff&#233;rencier les bact&#233;ries selon la composition de leur paroi cellulaire, les classant en bact&#233;ries gram-positives ou gram-n&#233;gatives. Cette technique commence par la pr&#233;paration d&#8217;un frottis bact&#233;rien fix&#233; &#224; la chaleur, qui est d&#8217;abord color&#233; au violet de gentiane. On ajoute ensuite de l&#8217;iode, formant un complexe violet de gentiane-iode qui se retrouve pi&#233;g&#233; dans la paroi cellulaire. Un traitement ult&#233;rieur avec un d&#233;colorant &#224; base d&#8217;alcool &#233;limine ce complexe chez les bact&#233;ries gram-n&#233;gatives, mais pas chez les gram-positives. Cette diff&#233;rence s&#8217;explique par l&#8217;&#233;paisse couche de peptidoglycane des bact&#233;ries gram-positives, qui retient la teinture violette. &#192; l&#8217;inverse, les bact&#233;ries gram-n&#233;gatives poss&#232;dent une couche plus mince de peptidoglycane ainsi qu&#8217;une membrane externe qui est alt&#233;r&#233;e par l&#8217;alcool, entra&#238;nant la perte de la coloration violette. La safranine est ensuite appliqu&#233;e en tant que contre-colorant, colorant les bact&#233;ries gram-n&#233;gatives en rose tandis que les gram-positives demeurent violettes.
Technique de coloration acido-r&#233;sistante
La coloration acido-r&#233;sistante est particuli&#232;rement utile pour identifier les esp&#232;ces du genre Mycobacterium, dont les parois cellulaires riches en acide mycolique r&#233;sistent aux colorations et d&#233;colorations conventionnelles. Dans cette proc&#233;dure, un colorant rouge, la fuchsine ph&#233;niqu&#233;e, est appliqu&#233; sur un frottis fix&#233; &#224; la chaleur, souvent avec une source de chaleur, pour faciliter la p&#233;n&#233;tration du colorant selon la m&#233;thode de Ziehl-Neelsen. Une solution d&#8217;alcool-acide est ensuite utilis&#233;e pour &#233;liminer la teinture des cellules non acido-r&#233;sistantes. Les bact&#233;ries acido-r&#233;sistantes, en raison de leurs parois cireuses, retiennent la coloration rouge, tandis que les cellules non acido-r&#233;sistantes sont contre-color&#233;es en bleu, g&#233;n&#233;ralement &#224; l&#8217;aide de bleu de m&#233;thyl&#232;ne. Il convient de noter que la m&#233;thode de Kinyoun, une variante de la coloration acido-r&#233;sistante, ne n&#233;cessite pas de chaleur pour l&#8217;application de la fuchsine.

Gram and acid-fast staining methods help scientists tell bacteria apart by looking at their cell walls. These differential stains are important in microbiology because they support identification, diagnosis, and research. They use structural and chemical differences in bacterial cell walls to separate microorganisms into clear groups.

Gram staining classifies bacteria as gram-positive or gram-negative. A heat-fixed bacterial smear is first stained with crystal violet, and iodine is added to form a crystal violet-iodine complex. Alcohol then acts as a decolorizer. It removes the complex from gram-negative cells but not from gram-positive cells.

The reason for this difference is the cell wall structure. Gram-positive bacteria have a thick peptidoglycan layer, which holds the purple stain. Gram-negative bacteria have a thinner peptidoglycan layer and an outer membrane. Alcohol disrupts that outer membrane, so the purple stain is lost. Safranin is then used as a counterstain. It turns gram-negative bacteria pink while gram-positive bacteria stay purple.

Acid-fast staining is used to identify Mycobacterium species. These bacteria have mycolic acid-rich cell walls, which resist normal staining and decolorization. Carbolfuchsin, a red dye, is applied to a heat-fixed smear. In the Ziehl-Neelsen method, heat is often used to help the dye enter the cell. Acid-alcohol then removes the stain from non-acid-fast cells.

Acid-fast bacteria keep the red stain because their waxy cell walls hold it. Non-acid-fast cells lose the red dye and are counterstained blue, usually with methylene blue. A related method, the Kinyoun stain, does not use heat for carbolfuchsin application. Both gram staining and acid-fast staining are essential for microbial identification, clinical diagnostics, and choosing the right treatment. Gram-negative bacteria can be more resistant to some antibiotics because of their outer membrane, so identifying them can guide therapy.
